Isolation of <em>Pseudocercospora griseola</em> (Sacc.) Ferraris (=<em>Isariopsis griseola</em> Sacc.), causing angular leaf spot on common bean grown in greenhouses in the Western Blacksea Region

Abstract

Bean has been an important choice for consumers because of its high protein rate and for Western Blacksea farmers because of its easy marketability. Pseudocercospora griseola, causing angular leaf spot on beans, which was firstly detected in Çaycuma district of Zonguldak province, Turkey, causes serious damage on local varieties (Ozan 2009).


The most important reason limiting studies on this fungus is its slow growth or not growing on artificial media. P. griseola is a slow-growing and difficult-isolated mitosporic fungus. Conventional mycological isolation methods were tried but it was not possible to grow the fungus. In this study, the most suitable isolation methods for this fungus were investigated by trying new methods and modified isolation methods used for some other fungi belonging to the same genus.

Abstract

Fasülye yüksek protein oranına sahip olması nedeniyle tüketicilerin, pazar sorununun olmamasından dolayı Batı Karadeniz Bölgesi üreticilerinin önemli tercihi olmuştur. Ülkemizde ilk defa Zonguldak ili Çaycuma ilçesinde tespit edilen ve fasülyede köşeli yaprak lekesine neden olan Pseudocercospora griseola bu bölgedeki yerel fasülye çeşitlerinde ciddi zararlara neden olmuştur (Ozan 2009).
Bu fungusla yapılan çalışmaları kısıtlayıcı önemli nedenlerden birisi yapay besi yerlerinde çok yavaş gelişmesi veya gelişmemesidir. P. griseola izolasyonu sırasında güçlük yaşanan ve çok yavaş gelişen mitosporik bir fungustur. Bu nedenle mikolojik açıdan bilinen klasik izolasyon metotları denenmiş ve etmen geliştirilememiştir. Bu çalışmada fungusun ait olduğu cinsteki diğer türlerin izolasyon yöntemleri, bu yöntemlerin modifiye edilmiş şekilleri ve daha önce denenmeyen bazı yöntemler denenerek etmen için en uygun izolasyon yöntemleri araştırılmıştır.
