Morphometric Assessment of Alveolar Bone Loss Following Systemic Darbepoetin Alfa Administration in a Rat Model of Ligature-Induced Periodontitis

Abstract

Aim: This experimental study investigated whether systemic administration of darbepoetin alfa could reduce alveolar bone loss in a rat model of ligature-induced periodontitis through morphometric evaluation. Materials and Methods: Fifty adult male Sprague–Dawley rats were randomly allocated to five experimental groups (n = 10): Control, Control receiving darbepoetin alfa (0.25 µg/kg/day), Periodontitis, Periodontitis treated with darbepoetin alfa (0.1 µg/kg/day), and Periodontitis treated with darbepoetin alfa (0.25 µg/kg/day). Experimental periodontitis was established by placing silk ligatures around the mandibular first molars. Darbepoetin alfa was administered intraperitoneally once daily throughout the 11-day experimental period. Following euthanasia, morphometric assessment of alveolar bone loss was performed by determining the linear distance between the cementoenamel junction and the alveolar bone crest using stereomicroscopic images. Results: Ligature placement resulted in a marked increase in alveolar bone loss compared with the healthy control groups (p < 0.05). Administration of darbepoetin alfa significantly limited bone loss in both treatment groups relative to untreated periodontitis (p < 0.05). Mean alveolar bone loss values were 0.42 mm in the Control group, 0.44 mm in the Control + darbepoetin alfa group, 1.27 mm in the Periodontitis group, 0.79 mm in the Periodontitis + 0.1 µg/kg/day group, and 0.77 mm in the Periodontitis + 0.25 µg/kg/day group. No statistically significant difference was identified between the two darbepoetin alfa dosage regimens (p > 0.05). Conclusion: Systemic administration of darbepoetin alfa significantly reduced alveolar bone loss in a ligature-induced rat model of periodontitis. Both treatment doses demonstrated comparable protective effects. Further experimental and clinical studies are needed to confirm these findings.
