Comparative Studies on the Diagnosis of Rabies in Cattle and Anatomical Localization of Rabies Virus by Real Time PCR

Abstract

Rabies is a fatal disease of all warm blooded animals. In this investigation the distribution of the rabies virus was determined by real time PCR, and comparison of PCR against histopathology, Seller reaction and florescent antibody test for diagnosis, was made. The material of the research was composed of 25 head of the cattle with rabies infection that was brought to Adana Veterinary Control and Research Institute between the years 2007-2009. All of the cases were determined viral nucleic acid by real time PCR. Eleven of them were positive by Seller, 15 were positive by histopathologically and 21 was positive by fluorescent antibody reaction for rabies. In this research it was observed that the amount of the viral nucleic acid was higher in cerebellum than the other parts of the head. A positive correlation was found between the viral load on the different areas of the head and viral load on origin of the nerves innerved these areas. The results of this study confirmed the real time PCR can be used in cases that rabies cannot be diagnosed by routine methods and absence of cerebrum and cerebellum.

Sığırlarda Kuduz Hastalığının Teşhisi ve Kuduz Virüsünün Anatomik Lokalizasyonunun Real Time PCR ile BelirlenmesiÜzerine Karşılaştırmalı Çalışmalar.

Abstract

Kuduz bütün sıcakkanlı canlılarda görülen, öldürücü zoonoz bir hastalıktır. Bu çalışmada Real Time PCR tekniği ile kuduz virüsünün sığır kafasındaki lokalizasyonları tespit edilerek, kuduz hastalığının teşhisinde Real Time PCR ile histopatoloji, Seller boyama ve floresan antikor testi (FAT) karşılaştırıldı. Çalışmanın materyalini Adana Veteriner Kontrol ve Araştırma Enstitüsüne 2007-2009 yılları arasında getirilen kuduz pozitif 25 adet sığır kafası oluşturdu. Real Time PCR ile kuduz olduğu belirlenen 25 olgunun 21’inde FAT ile 11’inde Seller boyama ile 15’inde histopatolojik olarak kuduza ait bulgular tespit edildi. Real Time PCR ile cerebellum’larda belirlenen viral nükleik asit miktarının diğer bölgelere göre daha fazla olduğu, kafanın farklı bölgelerinde ve bu bölgeleri inerve eden sinirlerin orijin yerlerinde belirlenen viral nükleik asit miktarları arasında pozitif korelâsyonun bulunduğu belirlendi. Beyin ve beyinciğin klasik yöntemlerle muayeneye uygun olmadığı durumlarda Real Time PCR'nun teşhis için kullanılabileceği ve yine beyin ve beyinciğin bulunamadığı durumlarda kafanın farklı bölgelerinden alınan örneklerden Real Time PCR ile kuduz hastalığının teşhisinin yapılabileceği kanaatine varıldı.
