Invitro Study of the Antimicrobial Effectives of Commercially Available Toothpastes

Abstract

Dental caries are formed by the increase in number in dental plate of such bacteriae Streptococcus mutans, Lactobacillus spp. The bacteriae produce acid by breaking down the carbohydrates very quickly. Therefore, one of the objectives of the caries preventive interventions is to inhibit the formation of the acid. Candida albicans causes aphthous lesions in the mouth and also can cause frequent infections in those who having dental prostheses. The main cause of periodontal dental caries is that the bacteriae and dental plaque and the solution is oral hygiene including teeth brushing. For this reason this study analyzes the effects of toothpastes in different brands and different contents on these bacteriae. Baskent University students randomly selected from among 169 people applied to the survey to determine the toothpates to be tested. According to the survey, invitro comparative antimicrobial effectiveness of thirteen selected commercially available toothpaste on Streptococcus mutans, Lactobacillus acidophilus, Candida albicans were investigated by using the agar well diffusion method. Thirteen toothpastes are numbered regardless of the number of toothpastes preferred. Chlorhexidine mouthwash as positive control, blank wells as negative control were used. Bacterial dilutions were prepared as recommended as antibiogram preparation and these mixtures inoculated into Mueller Hinton feed lots with and without blood via swap. Wells of a microtiter plate is opened and tested toothpaste is put 6 wells for the each . Toothpastes placed into the wells were weighed and inhibition zone diameters were measured waiting after 24 hours at 37° C. The data obtained were evaluated statistically. According to the zone diameters, compared to controls, the number 2 toothpaste to L.acidophilus and C.albicans and number 8 toothpaste to S.mutans has a lesser effect. As percent activity is evaluated mm/g none was effective as controls. The efficacy of toothpastes found to be lower than the positive control group. Accordingly the antimicrobial efficacy of toothpastes is not crucial. Mechanical brushing of the teeth is more critical to prevent tooth decay.

Ticari olarak satılan diş macunlarının antimikrobiyal etkinliğinin in vitro çalışması

Abstract

Diş çürüğü, diş plağında Streptococcus mutans, Lactobacillus spp. gibi mikroorganizmaların sayıca artmasıyla oluşur. Bu bakteriler karbonhidratları çok hızlı bir şekilde parçalayarak asit oluştururlar. Bu nedenle çürük önleyici girişimlerin amaçlarından biri de bu asit oluşumunu engellemektir. Candida albicans da ağızda aftöz lezyonlar yapmakta ayrıca diş protezi olanlarda sık enfeksiyona neden olabilmektedir. Diş çürüğü ve periodontal problem de neden bakteri yani dental plaktır, çözüm diş fırçalamak yani ağız hijyenidir. Bu nedenle bu çalışmada farklı marka ve içerikte diş macunlarının bu bakteriler üzerindeki etkisi araştırılmaktadır. Test edilecek diş macunlarını belirlemek için Başkent Üniversitesi öğrencileri arasından rasgele seçilen 169 kişiye anket uygulanmıştır. Anket sonuçlarına göre seçilen onüç ticari olarak satılan diş macununun in vitro olarak Streptococcus mutans, Lactobacillus acidophilus, Candida albicans üzerindeki antimikrobiyal etkinlikleri karşılaştırılmalı olarak agar well difüzyon yöntemi kullanılarak olarak incelenmiştir. 13 diş macunu tercih edilme sayısından bağımsız olarak rastgele numaralandırılmıştır. Pozitif kontrol olarak klorheksidin gargara, negatif kontrol olarak boş kuyucuk kullanılmiştır. Antibiyogram yapılırken önerilen şekilde bakteri sulandırımları hazırlanmış, bu karışımlar eküvyonla, hazırlanan Koyun kanlı ve kansız Müeller Hinton besiyerlerine inoküle edilmiştir. Kuyucuklar açılmış ve test edilen diş macunları kuyucuklara her biri için 6 kuyucuk olacak şekilde konulmuştur. Kuyucuklara konan diş macunlarının ağırlıkları tartılmış ve 37° C de 24 saat beklemenin ardından oluşan inhibisyon zon çapları ölçülmüştür. Elde edilen veriler istatistiksel olarak değerlendirildi. Zon çapları kontrolle karşılaştırıldıklarında, 2 numaralı diş macunu L.acidophilus ve C.albicans’a 8 numaralı diş macunu S. mutans’ a daha az etki yapmıştır. Yüzde etkinlik olarak değerlendirildiğinde mm/g ; hiçbirisi kontrol kadar etkili olmamıştır. Diş macunlarının etkinliği pozitif kontrol grubuna göre düşük bulunmuştur. Buna bağlı olarak diş macunlarının antimikrobiyal etkinliğine fazla anlam yüklememek gerekmektedir. Diş çürüklerine engel olmak için önemli olan dişin mekanik temizliğidir
