Detection of Cryptosporidiosis in Diarrhoeic Neonatal Calves in Nevşehir District by Real Time PCR and Nested PCR Techniques

Nevşehir Yöresindeki Yeni Doğan İshalli Buzağılarda Cryptosporidiosis’in Real Time PCR ve Nested PCR Yöntemleri ile Saptanması

Abstract

Bu çalışma, Nevşehir yöresindeki ishalli buzağılarda cryptosporidiosis’in real time PCR ve nested PCR teknikleri ile saptanması amacıyla yapılmıştır. Bu amaçla 2010-2011 yılları arasında Nevşehir’in farklı ilçe ve köylerinden 150 ishalli buzağıdan dışkı örneği tekniğine uygun olarak toplanmıştır. Toplanan örneklerden genomik DNA ekstraksiyonu yapılmıştır. Elde edilen genomik DNA’lar Cryptosporidium soy spesifik 18S rRNA parsiyel gen bölgesini amplifiye eden JVAF, JVAR primerleri ve JVAP TaqMan probuyla, C. parvum spesifik JVAGF, JVAGR primerleri ve JVAGP2 probu ile real time PCR analizine tabii tutulmuşlardır. Nested PCR analizlerinde de Cryptosporidium türlerinin 18S rRNA gen bölgesini amplifiye eden spesifik primerler kullanılmıştır. Real time PCR analizleri sonucu 23’ü (%15.3) C. parvum, 8‘i (%5.3) ise Cryptosporidium spp. olmak üzere toplam 31 (%20.7) örnekte cryptosporidiosis pozitifliği saptanmıştır. Örneklerin nested PCR analizleri sonucunda ise 29’u (%19.3) Cryptosporidium spp. pozitif bulunmuştur. Real time PCR tekniği altın standart test kabul edilerek nested PCR tekniğinin duyarlılığı %93.5 ve özgüllüğü ise %100 olarak belirlenmiş, iki teknik arasında %95.8 uyum saptanmıştır. Cryptosporidiosis’in en yüksek %23.2 ile 1-2 aylık buzağılarda belirlenmiş bunu sırasıyla %19.3 ve %16.7 ile 0-1 ve 2-3 aylık buzağılar izlemiştir. Enfeksiyonun erkek buzağılarda %21.5, dişilerde ise %19.7, Holştayn, Simental ve Montofon ırkında sırasıyla %32.6, %25 ve %11.1 yaygın olduğu görülmüştür. Bu çalışma Türkiye’de sığırlarda cryptosporidiosis’in real time PCR tekniği ile araştırıldığı ilk çalışmadır. Çalışmada Türkiye’de ilk kez Cryptosporidium soy ve C. parvum pozitifliği eşzamanlı olarak ortaya konmuştur
