Türkiye’de Yetiştirilen Anadolu Mandalarında Butirofilin (BTN1A1) Gen Polimorfizminin HaeIII Restriksiyon Enzimi ile Araştırılması

Abstract
Butirofilin süt yağının salgılanmasında önemli rolü olan
bir transmembran glikoproteinidir. Bu araştırmada, Türkiye’de yetiştirilen
Anadolu mandalarında butirofilin (BTN1A1) geninin genotip ve allel
frekanslarının belirlenmesi amaçlanmıştır. Bu çalışmada toplam olarak 120 manda
polimeraz zincir reaksiyonu-restriksiyon parçacık uzunluk polimorfizmi
(PCR-RFLP) metodu ile genotiplendirilmiştir. Çalışmada yapılan PCR işlemi sonunda
elde edilen 501 bç’lik amplikonlar elde edilmiştir. Elde edilen PCR ürünlerinin
HaeIII restiriksiyon enzimi ile
kesilmeleri sonucunda incelenen örneklerde sadece A alleli ve AA genotipinin bulunduğu
görülmüştür. Bu çalışma Anadolu mandalarında BTN1A1 gen polimorfizminin araştırıldığı
ilk çalışmadır. Çalışma sonunda, Anadolu mandalarında BTN1A1 geninde
polimorfizm bulunamamıştır.

Abstract
Butyrophilin is a
transmembrane glycoprotein and plays a critical role in secretion of milk
lipid. The aim of this study was to identify genotype and allele frequencies of
butyrophilin (BTN1A1) gene in Anatolian buffaloes in Turkey. A total of 120
buffalos were genotyped by the polymerase chain reaction-restriction fragment
length polymorphism (PCR-RFLP) method in this study. As a result of PCR
process, 501 bp amplicons were obtained from the Anatolian buffaloes examined
in this study. After digestion of the PCR products with HaeIII restriction enzyme, only A allele and AA genotype were
found. This study provided first information on the polymorphism on BTN1A1 gene
in Anatolian buffaloes. In conclusion, there was no polymorphism found in the BTN1A1
gene in Anatolian buffaloes.
