ÇEVRESEL OLARAK ASBESTE MARUZ KALAN KİŞİLERİN LENFOSİTLERİNDEKİ NÜKLEER BÖLÜNME İNDEKSİ (NDI) DEĞERLERİNİN İNCELENMESİ

Abstract

Asbest minerallerine maruziyet; akciğer kanseri, malign plevral mezotelyoma ve çeşitli organlardaki kanser vakalarını içeren ciddi sağlık etkilerinin geniş bir çeşitliliği ile ilişkilidir. Türkiye’de pek çok bölgede asbest bulunmaktadır. Bu bölgelerden biri olan Doğanlı Köyü’nde yaşayan ve hemen hemen doğduğundan beri asbeste maruz kalan kişilerin lenfositlerinde Nükleer Bölünme İndeksi (NDI) oranları değerlendirildi. Doğanlı Köyü’nde yaşayan, yaklaşık 30-70 yaşları arasında her iki cinsiyetten 30 hastanın periferik kanları alındı. Yaş, cinsiyet, ve sosyoekonomik bakımdan aynı olan Kayseri’ye bağlı bir köyden 25 kişi de kontrol grubu olarak kullanıldı. Alınan kan örnekleri, %25 fetal kalf serum ve fitohemaglutinin içeren Ham’s F-10 medyumlarına ekildi. 44. saatte Sitokalazin-B (Cyt-B) eklenerek, binükleer hücreler elde edildi. 72 saatlik kültür sonunda preparatlar rutin işlemlerle hazırlandı. Elde edilen preparatlar mikroskopta incelenerek, 1000 tane bir çekirdekli (mononükleer) hücre sayılırken aynı zamanda iki çekirdekli, üç çekirdekli, dört çekirdekli (binükleer, trinükleer ve tetranükleer) her bir hücre kaydedildi ve NDI değerleri hesaplandı. Bu çalışmada, asbeste maruz kalan kişilerin ve kontrol kişilerin NDI değerleri arasında fark bulundu (p=0.001). Sonuç olarak, her iki grup kişilerin NDI değerleri arasında anlamlı fark bulunması asbest maruziyetinin akciğer kanseri ve malign plevral mezotelyoma (MPM) ile olan ilişkisini desteklemektedir

Investigation of Uclear Division Index (NDI) in lymphocytes of People who are Environmentally Exposed to Asbestos

Abstract

Exposure to asbestos minerals has been associated with a wide variety of adverse health effects including lung cancer, malign pleural mesothelioma, and cancer of other organs. There are asbestos deposits in many regions of Turkey. People in the village of Doğanlı (one of those regions) have been exposed to asbestos all their lives. These people’s Nuclear Division Index (NDI) rations in their lymphocytes were evaluated. Peripheral blood samples were taken from 30 subjects who live in Doğanlı village, age range 30-70, from each gender. Twenty-five people of the same age, sex and socioeconomic group from a village near Kayseri were used as a control group. Blood samples were cultured in Ham’s F-10 medium, which consists of 25% fetal calf serum and phytohemaglutinin. At 44th hour for binucleated cells, Cytochalasin-B (Cyt-B) was added to cultures. After incubation of 72 hours, the slides were prepared according to routine methods. One thousand mononucleated cells were scored for the NDI rations by microscope on slides. At the same time each cell that has two nucleus, three nucleus and four nucleus (binucleated, trinucleated and tetranucleated) were scored and the NDI rations were calculated. In this study, there were significant differences between the NDI values of people who were exposed to asbestos and people in the control group (p=0.001). As a result, these significant differences between the NDI values support that exposure to asbestos is associated with lung cancer and malign pleural mesothelioma (MPM)
