SIÇANLARDA SİGARA İNHALASYONU SONUCU PREFRONTAL KORTEKSTE OLUŞAN YAPISAL DEĞİŞİKLİKLER ÜZERİNE KAFEİK ASİT FENETİL ESTER (CAPE)’İN ETKİSİ

Abstract

Bu çalışmada, sigaranın prefrontal korteks üzerine olan nörotoksik etkileri ve bu toksik etkilere karşı kafeik asit fenetil ester (CAPE)’in koruyucu etkisi ışık mikroskobik düzeyde araştırıldı. Bu amaçla 21 adet Wistar cinsi erkek sıçan üç gruba ayrıldı. Grup I’deki hayvanlar kontrol grubu olarak kullanıldı. Grup II’deki sıçanlar sigara dumanına maruz bırakıldı. Grup III’deki sıçanlara ise sigaraya maruziyeti ile birlikte günlük olarak CAPE enjekte edildi. 60 günlük deney süresi sonunda tüm sıçanlar dekapitasyonla öldürüldü. Daha sonra sıçanların beyinleri çıkarıldı. % 10’luk formaldehit ile tespit edilen prefrontal korteks doku örnekleri rutin histolojik prosedürleri takiben ışık mikroskobunda incelendi. Sigara dumanına maruz bırakılan sıçanlara ait prefrontal korteks örneklerinde, moleküler tabakada vakuolizasyon, nöroglial hücre infiltrasyonu ve piknotik hücreler tespit edildi. Buna karşın, sigara maruziyeti ile birlikte CAPE enjekte edilen sıçanlarda ise, sigara maruziyetinin neden olduğu yapısal değişikliklerin büyük oranda azaldığı, ancak sigara maruziyetine benzer şekilde piknotik hücrelerin varlığını devam ettirdiği belirlendi. Sonuçta, sıçanlarda sigara dumanı maruziyetinin prefrontal kortekste hasara neden olduğu ve bu hasarın CAPE uygulaması ile önlendiği tespit edildi

The Effects of Caffeic Acid Phenetyl Ester (CAPE) on Structural Changes in the Prefrontal Cortex Induced by Cigarette Inhalation in Rats

Abstract

In this study, neurotoxic effects of cigarette on prefrontal cortex and protective effects of caffeic acid phenetyl ester (CAPE) against these toxic effects were investigated at light microscopic level. For this purpose, 21 male Wistar rats were divided into three groups. Animals in Group I were used as control. Rats in Group II were exposed to cigarette smoke. The rats in Group III were exposed to cigarette smoke and injected daily with CAPE. At the end of the 60-days experimental period, all rats were killed by decapitation. Then the brains of rats were removed. Following routine histological procedures, the prefrontal cortex tissue specimens fixed in 10% formaldehyde were examined under a light microscope. Light microscopic examination of prefrontal cortex specimens from rats exposed to cigarette smoke revealed that vacuolization, neuroglial cell infiltration and pycnotic cells in the molecular layer. Whereas, exposure of cigarette smokeinduced structural changes were decreased in rats exposed to cigarette smoke and injected with CAPE, but only pycnotic cells were seen similar to the cigarette smoke group. In conclusion, It was determined that exposure to cigarette smoke caused damage in prefrontal cortex of rats and this damage was prevented by administration of CAPE
