Molecular Characterozation of Some Free-Living Amoeba in Soil Which Can Be Parasitising in Human

Abstract

Acanthamoeba, Naegleria, Balamuthia, Sappinia, Vahlkampfia and Hartmannella of the free-living ameba are commonly encountered in soil, air, lens solutions and water sources such as tap water, lakes, rivers and swimming pools. Some of those free-living ameba may cause serious diseases in human. The main objective of this study is to isolate and genotype the free-living potential pathogen amoeba species in the soil samples obtained from different regions of Kayseri. Each of the total 20 samples are passed through cellulose nitrate filter and transported to the non nutritional plates and coated with Escherichia coli agar. The samples which are cultured at 37 0C are observed for two weeks in order to observe the reproduction and isolate the DNA samples that are thought to be positive. Eight of the 20 samples which are undergoing PCR with specific primers were Acanthamoeba positive and three of them were Naegleria positive. Besides, Balamuthia was not detected in any of the samples. In conclusion, in this study the presence of some free-living amoeba species which may threaten human health is shown in the soil samples obtained from Kayseri
