KİSTİK EKİNOKOKKOZİSLİ HASTALARDA OKSİDATİF STRES: OKSİDAN VE ANTİOKSİDAN PARAMETRELER ARASINDAKİ İLİŞKİ

Abstract

Kistik ekinokokkozis (KE), dünyada ve Türkiye’de insan sağlığını tehdit eden paraziter bir zoonozdur. Oksidatif stres oksidan üretimi ile antioksidan korunma arasındaki dengenin bozulması nedeniyle ortaya çıkar ve protein, lipid ve nükleik asitlerin oksidatif modifikasyonu nedeniyle yapısal hasarla sonuçlanabilir. Bu çalışmada, KE’li bireylerde serbest radikal oluşumu ile antioksidan savunma mekanizmaları arasındaki ilişki ve sonuçta doku hasarının derecesinin incelemesi amaçlandı. Çalışmaya 23 hasta ve 25 kontrol birey dahil edildi. Malondialdehid (MDA) düzeyi, ksantin oksidaz (XO) ve katalaz (CAT) aktiviteleri spektrofotometrik yöntemlerle ölçüldü. KE’li grupta kontrol grubuna göre MDA düzeyi ve CAT aktivitesi rakamsal olarak oldukça yüksek bulunurken bu fark istatistiksel olarak anlamlı bulunmadı, (p= 0,32, p= 0, 11). Buna karşılık XO aktivitesi hasta grubunda kontrol grubuna göre istatistiksel olarak anlamlı düzeyde yüksek bulundu (p= 0,0023). Yapılan korelasyon analizleri ve istatistiksel değerlendirme sonucunda XO aktivitesi ile MDA düzeyi arasında anlamlı bir negatif korelasyon görüldü (p< 0,0001). CAT aktivitesi ile MDA düzeyi arasında aynı şekilde anlamlı negatif korelasyon görülürken (p= 0, 0014), CAT ve XO aktiviteleri arasında anlamlı pozitif korelasyon tespit edildi (p= 0,0014). Elde edilen bu sonuçlar özellikle parazit kaynaklı patolojilerin neticesinde organizmada oluşabilecek doku hasarının derecesi hakkında bilgi verip, verilecek tedavinin nitel ve nicelik olarak yönlendirilmesi aşamasında faydalı olabilecektir

Oxidative Stres in Patients with Cystic Echninococcosis: Relationship Between Oxidant and Antioxidant Parameters

Abstract

Cystic echinococcosis (CE) is a parasitic zoonose which threatens human health in the world and Turkey. Oxidative stress occurs due to imbalance between oxidant production, and antioxidant protection and may result in structural tissue damage due to oxidative modification of protein, lipid and nucleic acids. In this study, we aimed to investigate free radical formation and its relationship between antioxidant mechanisms and thus tissue damage degree in patients with CE. Twenty-three patients and 25 controls were included in the study. (Malondiladehyde) MDA levels, xantine oxidase (XO) and Catalase (CAT) activities were measured using spectrophotometric method. MDA and CAT levels were found to be numerically higher in CE patients compared to controls, but this difference was not statistically significant (p= 0,32, p= 0, 11). However, XO activity was found to be statistically significant in patients compared to the controls (p= 0,0023). Correlation analysis and statistical evaluation together showed that there was a significant negative correlation between XO activity and MDA level (p< 0,0001) and also significant negative correlation between CAT activity and MDA level (p= 0, 0014), however, there was a significant positive correlation between CAT and XO activities (p= 0.0014). In conclusion, these results will provide some more information relating to the degree of tissue pathology as a result of parasite infection, and accordingly it would be also useful to give direction to the treatment in qualitative and quantitative degrees
