KAYSERİ YÖRESİNDE SÜT SIĞIRLARINDA ESCHERICHIA COLI O157:H7’NİN KONVANSİYONEL VE MOLEKÜLER YÖNTEMLERLE ARAŞTIRILMASI

Abstract

Bu çalışmada, Kayseri ili ve ilçelerindeki kamu ve özel sektöre ait süt işletmelerindeki çeşitli yaş gruplarındaki sağlıklı hayvanların dışkı ve sütlerinde E. coli O157:H7’nin varlığının saptanması ve hayvansal risk faktörlerinin ortaya konulması amaçlanmıştır. Bu amaçla çeşitli ilçelerden toplanan 500 adet dışkı ve 500 adet süt örnekleri çalışmada materyal olarak kullanıldı. Immuno Magnetik Seperasyon ve ardından Chorom agar ve Sorbitol MacConkey besiyerlerinde üreyen şüpheli koloniler seçildi ve bunlar serolojik yönden H ve O agglütinasyon testi ile incelendi. Moleküler yönden ise şüpheli izolatlar multipleks PCR (mPCR) ve unipleks PCR (uPCR) teknikleri ile incelendi. Bu işlemleri sonucunda 500 süt örneğinden 1 (% 2)’inde ve 500 dışkı örneğinden 6 (% 4,6)’sında O157:NM suşu tespit edilmiş olup örneklerin hiçbirinde flagella H antijenine rastlanmadı. Dışkıdan izole edilen O157:NM suşlarına yapılan mPCR’ da bir örnekte Enterohemolysine genine (Ehly A) rastlanırken, 1 örnekte yalnızca Sitotoksin 2 (stx2) geni tespit edildi. Diğer dışkı örneklerinde virülens genlerinden hiçbirisine rastlanmadı. Sütten izole edilen bir adet O157:NM suşunda ise virülens genlerinden hiçbirisine rastlanmadı. Sonuç olarak, Kayseri ili ve ilçelerindeki işletmelerdeki süt sığırlarının dışkısında ve sütlerinde E. coli O157 belirlenmiştir. E.coli O157:H7 tespit edilmemiştir. Belirlenen kontaminasyon oranının düşük düzeylerde olduğunu, bununla birlikte Kayseri ilinde hijyen kurallarına dikkat edilmesi gerektiği sonucuna varıldı

Investigation with Conventional and Molecular Method of Escherichia Coli O157:H7 in Dairy Cattle in Kayseri Region

Abstract

The objective of the present study is to investigate the animal risk factors related to the presence of E. coli O157:H7 in feces and milk samples collected from healthy cattle at differing ages in private and government dairy farms in Kayseri and its vicinity. Five hundred fecal samples and 500 raw milk samples were collected from different locations in Kayseri, processed through immune Magnetic Separation, and inoculated Chorom medium and Sorbitol MacConkey medium. Suspicious colonies were selected from this medium, and they were examined serologically with H and O agglutination test. The molecular direction suspicious isolates were examined using multiplex with PCR (mPCR) and uniplex PCR (uPCR) techniques. The examination showed that only 1 (2 %) out of 500 raw milk samples and only 6 (4.6%) out of 500 fecal samples contained O157:NM strain, and none of the samples contained flagella H antigen. Through using mPCR, only one sample from O157:NM of isolated fecal samples contained Ehly A, whereas only one sample contained stx2 genes. None of the other fecal samples contained any virulence genes. Only one raw milk sample isolated from O157:NM strain did not have any virulence genes.In conclusion, the raw milk and fecal samples collected from dairy farms in Kayseri region and its vicinity contain E. coli O157. Low levels of contamination rate was determined in the samples. However, it was concluded that due attention should be paid to hygiene rules in Kayseri
