SİYANÜR ve TİYOSİYANATIN TOKSİKOKİNETİĞİ ÜZERİNE SODYUM NİTRİTİN ETKİLERİNİN ARAŞTIRILMASI

Abstract

Bu çalışma, amigdalin ile zehirlenen koyunlarda kan siyanur ve tiyosiyana düzeylerine aiı bazı oksikokinetik parametreler uzerine sodyum nitritin etkisini araştırmak amacıyla gerçekleştirildi Bu çalışmada, ağııiıkiarı 50-60 kg arasında olan 18 koyun kullanıldı. Amigdalin ve sodyum nitritin verilmesinden sonra 0.5, 1, 2, 4, 8, 24, 48 ve 72. sa­ atlerde alınan kan örneklerindeki siyanür ve tiyosiyanat düzeyleri ile bunlara ait bazı toksikokinetik parametreler be­ lirlendi. Sodyum nitritin 5 ve 25 mglkg dozlarında damar içi yolla uygulanmasından sonra alınan kan siyanür düzeylerine ait dağılım dönemi yarı ömürlerinin dozlara göre sırasıyla, 0.23±O.01 ve 3.14±1.6S saat; atılım dönemi yarı ömulierinin sırasıyla, 60.80±13.41 ve 126±24.33 saat; ortalama kalış sürelerinin sırasıyla, 88±10.04 ve 18S±2S.21 saat; doruk yogunluğa ulaşma sürelerinin 48. saat; doruk siyanür düzeylerinin sırasıyla, 0.151 ve 0.148 �g1ml olduğu tespit edildi. Bunun yanında, siyanür düzeylerine ait eğri altında kalan alan değerleri sırasıyla, 11.28±1.24 ve 9±S.62 �g saaVml; kararlı durumdaki dağılım hacimleri .sırasıyla, 0.03±O.OOI ve 0.24±O.03 Ukg; klirens değerleri ise sırasıyla, 0.44±O.12 ve 0.16±O.09mVsaat kg olarak hesaplandı. Kan tiyosiyana düzeylerine ait dağılım dönemi yarı ömürlerinin dozlara göre sırasıyla, 18.30±2.21 ve 29.44±3.41 saat; atılım dönemi yan ömürlerinin sırasıyla, 224.43±t 1.40 va 42.89±8.8S saat; ortalama kalış sürelerinin sırasıyla, t 3 7.36±24.2 va 64.38±6.6S saat; doruk yoğunluğa ulaşma surelerinin sırasıyla, 24 ve 48. saat; doruk tiyosiyanaı duzeylerinin ise sırasıyla, 0.026 ve 0.028 �g1ml olduğu belirlendi. Ayrıca, tiyosiyanaı duzeylerinin eğri altında kalan·alan değerleri sırasıyla, 4.66±O.21 ve 9.82±0.8 �g saaVml; kararlı du­ rumdaki dağılım hacimleri sırasıyla, 0.147±O.03 ve 0.162±O.03 Ukg; klirans değerlerinin ise sırasıyla, 1.07±O.31 ve 7.72±2.84 ml/saat. kg olarak tespit edildi

The Investigation of Sodium Nitrite Effects on The Toxicokinetics of Cyanide and Thiocyanate

Abstract

This study was carried out to determine the effects of sodium nitrites on some toxicoklnetic parameters ol eyanide and thiocyanate in sheep experimentally poisoned with amygdalin In this study, 18 sheep 50 - 60 kg weights were used. Amygdalin and sodium nitrite were given and Ihe eyanide and thi�yanate levels in samples were de­lermined Then some toxicokinetic parameters ol the eyanide and thiocyanate in the blood samples taken at 0.5, 1,2, 4, 8, 24, 48 and 72 th hours were established. The sodium nitrite was given intravenously in doses 5 and 25 mglkg and then the distribution hall-lives of blood eyanide levels according to doses were respectively, 0.23±O.01 and 3.14±1.65 hours; the elimination hall-lives were respectively, 60.80±13.41 and 126±24.33 hours; mean residence times were respectively, 88±10.04 and 18S±2S.21 hours; the duration ol maximal concentration were respectively, 48th hour; the maximal blood levels were respectively, 0.151 and 0148 �g1ml were determined. Besides, area under the curve values were according to doses were respectively, 11.28±1.24 and 9±S.62 �g.hour'ml; volume of dist­ribution at steady slate were respectively, 0.03±O.OOI and 0.2±0.03 Ukg; clearance values were respectively, 0.44±O.12 and 0.16±O.09 ml/hour.kg were calculated. The distribution half-lives of blood thiocyanaıe levels according lo doses were respectively, 18.30±2.21 and 29.44±3.41 hours; the elimination half-lives were respectively, 224.43±11.40 and 42.89±8.85 hours; mean residence times were respectively, 137.36±24.2 and 64.38±6.6S hours; the duration of maximal concentration were respectively, 24 and 48th hours; the maximal blood levels were res­pectively, 0.026 and 0.028 �g1ml were detennined. Besides, area under the curve values according lo doses were respectively, 4.66±O.21 and 9.82±O.8 �g.hour/ml; volume ol distribution at steady state were respectively, 0.147±O.03 and 0.162±O.03 Ukg; clearance values were respectively, 1.07±O.31 and 7.72±2.84 mVhour.kg were established
