FARKLI TESPİT SIVILARININ SIĞIR, KOYUN VE KEÇİ PARATİROİD BEZLERİNİN İNCE YAPISI ÜZERİNE ETKİSİ

Abstract

Bu çalışmada, sığır, koyun ve keçi paratirold dokuları immersiyon yöntemiyle% 2.5 glutaraldehid (GA) veya% 1 glutaraldehid ve% 2 formaldehid (FA) ve bunu takiben % 1'1ik osmik asitte (Os04:0smium tetroxide) tespit edilerek; bu dokuların ince yapısını en iyi koruyan fiksasyon ve tampon çözeltilerin belirlenmesi amaçlandı. Araştırmada kul­lanılan bütün hayvan türlerinin paratirold dokularında üniform (ince yapısı optimal seviyede korunmuş hücre), açık (açık sitoplazmalı ve hücre organelleri fiksasyondan farklı derecede zarar görmüş hücre) ve koyu (koyu sitoplazmalı ve büzüşmüş yapıda olan hücre) hücreler gözlendi. Na/K-fosfat, Na-cacodylat veya Hepes [4·(2-Hydroxyethyl)-1· Piperazinethan-Sulfonic acid] varlığında aldehidlerle ve Na/K-fosfat veya Na-cacodylat'ın varlığında osmik asit ile tespit edilen paratirold dokusu örneklerinde, genellikle üniform ve az sayıda da açık ve koyu hücreler görüldü. Ancak Hepes tampon çözeltisiyle hazırlanan osmik asit fiksasyonunda, bütün preparatlarda açık hücrelerin sayısının arttığı gözlendi. Aynı işlem koyunlarda açık hücre sayısına ilave olarak multinükleer hücre (hücre zarının bütünlüğü bozulmuş ve or· ganelleri şiddetli derecede dilale olmuş hücre) sayısında da artışa neden oldu. Glutaraldehid-formaldehidile tespit edi­len dokularda membranların, sadece glutaraldehidile tespit edilenlere kıyasla daha iyi korunduğu ve membran bü­tünlüğü ile organelierin ince yapısında önemli kayıpların oluşmadığı gözlendi. Bu çalışmada tesbit edilen 4 farklı hücre tipinin (üniform. açık, koyu ve multinükleer) farklı fiksatif ve tampon çözeliilere bağlı olarak oluştuğu sonucuna-varıldı. Bu hücre tiplerinin ortaya çıkışında tampon solüsyonları nın. aldehidlerden daha önemli bir rol oynadığı belirlendi

The Effect of Different Fixation Media on the Fine Structure of Parathyroid Glands in Cattle, Sheep and Goat

Abstract

Tissue samples of parathyroid glands obtained from cattle, sheep and goat were immersed in aither 2.5% glutaraldehyde (GA), or in 1% glutaraldehyde and 2% formaldehyde (FA) followed by immersion in 1% osmium tet­roxide (Os04). The effect of different bulters used for aldehydes and osmium tetroxide respectively, were tested in order to determine the optimal conditions for preservation of parathyroid cell morphology. Uniform (the subcellular components of these cells showed no damage due to fixation), light (light cytoplasmic matrix and the subcellular com­ponents were damaged to a variable extent due to fixation) and dark (dark cytoplasmic matrix and shrunken cell) cells were found in the parathyroid tissues of all animals. Parathyroid tissue fixed with aldehydes in the presence of Na/K­phosphate, Na-cacodylate or Hep es [ 4-(2-Hydroxyethyi)-1-Piperazinethan-Sulfonic acid] (with or without Ca2+ and Mg2+). and with osmium tetroxide in Na/K-phosphate or Na-cacodylate contained predominantly uniform. few light and dark cells. However, number of light cells lncreased in parathyroid tissue fixed with osmlum tetroxlde In Hepes. In addition to the increased light cells, number of multinuclear ce lls (the cells contained fragments of the plasma memb­rane and markedly dilated cisternae of the rough endoplasmic reticulum) also increased in parathyroid tlssues of sheep. The structure is batter preserved and particularly the membranes are batter visible in preparations fixed with a mixture of glutaraldehyde and formaldehyde than in those immersed only in glutaraldehyde. lt is concluded that the appearance of tour different cell types depended to a great extent on the bulters and 1 or fixatives used in the ex­periment. lt is also concludet that bulters played more important roles then fixatives in the occurrence of these cell types
