The routine work in the Rabies Laboratory

Abstract

This article is a summary of the routine works of the Rabies Laboratory
at the Etlik Veterinary Hacteriology Institute.
1 - The samples that the are sent to be diagnosed from different
places in the country come in different forms and are subject to a
method of examination according to the form they come in.
a) If the head of the animal suspected of rabies comes, or pieces
of its brain in water containing 50 % glycerine, smears of thin slides
composed of Ammon's horn are made, and stained with Seller's
dye, and Negri body are examined under the microscope. It is possible
through this method, to arrive at a result in a very short time.
If the diagnosis could not be made with this method, the samples taken
from different parts of the brain suspected of disease are emulsified
in salt water and 10 % suspension is made. The suspension is inoculated
into the 3 to 4 week - old mice. At least five rnice are used for
each sample.
The mice are kept under observation for a month. The diagnosis
can be made certain by making smears from the brains of the mice
that died during the above mentioned timen and examining them under
the microscope.
b) It is not possible to obtain a particular result from the misroscopic
examination, if there is any putrefaction in the sample. In
this case, the 10 % suspension made from the brain with the addition
of antibiotics according to the degree of putrefaction is ~noculated
into at least ten mice.
c) If the sample has come in some kind of antiseptic, the pieces
taken form the Ammon's horn are cut in microtom and stained
with Lentz method.
2 - The rabies vaccine prepared by the Sample method using
Pasteur's Virus Fix is produced for the Veterinary Organization. The
brain of either a sheep or a goat is used. The vaccine is the 10 % sus·
pension of the brain made with salt water containing 0.5 % phenol.
The vaccine that is prepared, after being tested for its sterility, inocuty
and immunity is preserved at + 4 c degree.
For preventive purposes, dogs of normal size are vaccinated twice
with a dosage of 10 ml. and small dogs and cats are vaccinated twice
with a dosage of 5 ml. with a 10 - 14 days interval between the two vaccination.

In our laboratory, the vaccine prepared by avianized Kelev strain
is being worked upon.
The first table shows, with percentages, the number of the sampies
examined in our laboratory during the years 1957 - 1962, and the
positive cases according to the kind of animals.
The second table shows, with percentages, the number of samples
subjected to the inoculation of mice, and the positive cases.
Production of vaccine has also been shown in dosage over the
years.

Bu yazıda kuduz laboratuvarının günlük çalışma mevzuu olan marazi
madde muayenesi ve aşı istihsal metodları kısaca anlatılmış, ı957 -
ı962 senelerinde muayene edilen marazi madde adedi, müsbet çıkan
vak'alar, deneme inokülasyonu ve aşı istihsali doz olarak kaydedilmiştir.
