Abstract

Work on a small scale has been done regarding virus productin
by Frenkel metıhod which is more practical and sheaper, parallel
with other measures against foot and mouth. epidemics in Turkey
Adaptaıtiori inıto exp'ante cattle tongue using SAT - 1 virus in Roux
bottles. Satidacıtory results were obtained. Experiments indicate future on lorge scale by tıhis method depends on getiing fresh caıtıtle tonıgue as well as chemicals and aither materials. Modified Flrenkel and Brescia media were used in experiments and fallawing eacıh .paseaıge complement fixation tests by virus on animals were carried out, ca'culating titrages in mice which were DL₅₀ 10^-65

Complement fixation tests have confirmed SAT- 1 all the time.
Working, in principle, witıh fresh tongue epithelium rather than
conserving epithelium obtained from tongues, tongue epithelia produced were used .the same day.
SAT- 1 virususedin adaptation isa nrutural virus of bovine or'ıgin.
Two methods were used in these tests :
1- With agitation
2 - Without agitation
1 - İn the agi.t~tion metıhod, cuılture was slowly agitated iii1to
the magnetic system while continuing with oxy.gen circulation.
2 - İn the. second metıhod tıhe culture was placed in a slanting
position in Roux bottles and oxygen was circulated. This has conrtınued in 18 - 24 hours interva1.s in 37°C. Results compared did notindicate large differences.
Testıs were carried out up to ninth passage 'but viruses were at
their best at. sixth and seventh passages from both infectivity and
antigeniti points of view.

Frenkel Metodu ile Şap Virusu Üretilmesi

Abstract

Türkiye'deki şap salgınlarını diğer ted!b'rlere para!el olarak aşı- ·
!ama ile önlemek için, ekonomik ve pratik ·kolaylıklan bulunan
Frenkel Metodu ile şap virusu üretilmesi için, küçük çapta ,çalışma
yapılmıştır. SAT, virusu ile Ajitasyonlu ve Roux şişelerinde olmak
üzere explante sığır diJ.ıine adaptasyon yapılmıştır. Bu çalışmalardan
arzuya uygun sonuçlar alınmıştır. Denemelerimiz bize göstermiştir
ki, ilerideki geniş çalışmalarda şap virusunun bu metodla çoğaltılması,
malzeme ve şimik maddelerle taze sığır dilinin temininC'
bağlıdır. Denemelerde ModWe Frehkel, Brescia vasatları kullan:J,
mış, her pasajdan sonra virusun hayvan deneyleri ile Komp'ement
Fiksasyon testleri yapılıp, faredeki titrajları hesaplanmıştır. Faredeki
DL·• 10-6 , 5 bulunmuştur. · C.F. testlerinde daima SATı teyid
edilmiştir.
Dillerden elde edilen epitelierin muhafazasından çok, taze dil
epiteli ile çalışmak prensip edilip, istihsal edilen dil epitelleri aynı
gün kullanı1mıştır. Adaptasyonda kullanılan SAT, virusu sığır orijinli
trubii virustur. Bu çalışmalarda iki usul izlenmiştir.
1 - Ajitasyonlu,
2 - Ajitasyonsuz.
1 - Ajitasnoylu usulde manyetik sistemle kültür ağır ağır ajite
edilmi5, bu arada oksijen sirkulasyonu da devam etmiştir.

2 - Ajitasyonsuz usulde Roux şişelerinde yatık olarak kültüre
konulmuş ve yirie oksijen sevkedilmiş, bunlar 37°C lik etüvde
ı8 - 24 saat · fasılalarla tutulmuş, neticeler mukayese edilmiş, arada
büyük bir fark bulunamamıştır.
Denemeler 9 ncu pasaja kadar yürütülmüş, ancak gerek enfektivite,
gerekse antijenite yönünden 6 ve 7 nci pasaj viruslannın en
iyi durumda bulunduğu tesbit edilmiştir.
