Sutudies for Developing a Vaccine Against Vibrio Fetus in Sheep

Abstract

This ·stüdy was conducted in the years of 1973- 1976, with the object of determining the incidence of vibrio fetus and producing a vaccine against vibriosis in Turkey.
1 - 206 fetal materials (liver, spleen, brain and gastric Juice)
belonging to the aborted fetus of sheep from differents parts of Turkey (see. the map) were examined bacteriologically for vibriosis, and 31 native. vibrio strains were isolated.
2 - All of the vi brio strains reacted po si tively by serologically
with Serotype (Ol) of vibrio strains.
3 - When the vibrio strains kept in liver broth at room temperature,the stability of growth did not change at last four years,without af.i.y passages.
4 - These vibrio strains were found as pathogenic .experimentally for pregnant sheep as follows :
a) Pregnant sheep were aborted in soinedays after infection
with vibrio strains oraily.

b) Sero type Ol of the vibrio strains were reisolated again in
the 60 percent of fetal materials obtained from abortions of sheep.
5 - 1063 ·blood sera taken from aborted sheep from different
parts of Turh!y ex~mined for vibriosis by Complement fixation test,
and 468 (44 percent) of these sera we're foLLnd positive for vibriosis
(seet the map).
6 - Two kind of vaccines as being of' an aliminium hydroxided
live vaccine and an aliminium hydroxided vaccine inactivated
by formalin were produced with the native vibrio strains, isolated ' in the study.
Both live and inactive vaccines gave en excelleut immunty in
pregnant sheep, 100 percent and 91.7' percent respectively: It was also observed "that the immunity lasted in the second pregnancy of sheep vaccinated for ones in the experiments.
7 - Two kind of the vaccines were found harmlass for sheep
in safety tests. The both vaccines were used in 420 sheep as field experiment and there were no complications with them They gave also normal birth.
8 - A map was drawn first time to show where the infection
of sheep seJ·ologicaly and bacteriologicaly in Turkey. ,

Koyunlarda Vibrio Fetüsten İleri Gelen Sıkıtlara Karşı Bir Aşı Geliştirme Çalışmaları

Türkiye Bilimsel ve Teknik Araştırma Ki.ımmun'ca destekl~hıniş
olan bu araştırmamız 1973- 1976 yılları içerisinde gerçekleştirilmiştir.
1 - Yurdumuzun değişik bölgelerinde koyun sürükri ,arasmda
çıkan enfeksiyöz tabiattaki yavru atma vak'alarından, temin edilip
206 adet fetüs marazi maddesinden yapılan bakteriyolojik ekimler
sonuncin 31 vibrio suşu izole edilmiştir .
a) Ayırımı yapılan bu 31 vibrio suşunun vibrio- fetüs serotip:
or gurubuna ait oldukları deneysel olarak kanıtlanmıştır.
b) Yine bu vibrio suşlarının tümünün gebe koyunlar için patojen
oldukları isbatlanmıştır.
c) Suşlar karaciğerli buyyonda hiçbir pasaja tabi tutulmadan
oda sıcaklığında muhafaza edildiklerinde üreme yeteneklerini en az
dört yıl korudular.
2 - Yurdun değişik yörelerinden yavru atan ana koyunlardan
temin edilen 1063 kan serumu seroljik olarak komplement- fixasyon
testiyle işlenmiş olup bunlardan 468'inin vibriosis yönünden
reaktör oldukları saptanmıştır.
a) Muayeneye tabi tutulan bu serumlarda vibrio- fetüs serotip:
Ol e karşı teşekkül etmiş spesifik antikorların varlığı tesbit edilmiştir.
3 - Yerli vibrio fetüs suşlarından iki çeşit Alüminyumhidroksitli
aşı hazırlanmıştır.
a) Alüminyumhidroksitli canlı,
b) Alüminyumhidroksitli formolle inaktiv vibrio aşısı.
Deneme sonunda canlı aşıdan koyunlarda 0/cı 100 ve inaktiv
aşıdan ise % 91.7 oranda bir bağışıklık elde edilmiştir .
4 - Koyunlarda aşılardan ötürü herhangi bir kamplikasyon
görülmemiştir.
5 - Her iki aşı ile 420 koyun üzerinde saha tatbikatı yapılmış
olup tüm aşılılar normal doğum yapmışlar ve aşılardan ötürü hiç
bir komplikasyona rastlanmamıştır.
6 - Bu çalışınarnızla vibriosise karşı bir kere aşılanmış olan
koyunlarda bağışıklığın ikinci gebeliklerinde de devam ettiği deneysel
olarak kanıtlanmıştır.
