Klorprifos’un ratlarda ince bağırsak üzerine etkisi ve kuersetin ve kateşin’in koruyucu rolü

Abstract

Bu çalışmada organofosfatlı bir pestisit olan ve yaygın olarak kullanılan klorprifos’un ratların ince bağırsakları üzerine olumsuz etkileri ve kuersetin ve kateşin’in bağırsakları koruyucu özelliği araştırıldı. Bu amaçla erkek ratlara klorprifos (5.4 mg/kg 1/25 LD50), kateşin (20 mg/kg), kuersetin (20 mg/kg), kateşin(20 mg/kg) + klorprifos (5.4 mg/kg 1/25 LD50) ve kuersetin (20 mg/kg) + klorprifos (5.4 mg/kg 1/25 LD50) kombinasyonları oral olarak dört hafta süre ile uygulandı. Uygulamadan dört hafta sonra ratların ince bağırsak dokuları histopatolojik incelemeler için alındı ve ışık mikroskobunda kontrol grubu ile kıyaslamalı olarak incelendi. Kateşin ve kuersetin uygulanan gruplar ile kontrol grubu ratların ince bağırsak dokularında histolojik olarak farklılık gözlenmedi. Klorprifos uygulanan ratların ince bağırsak dokusunda villuslarda atrofi, nekroz ve muskular mukozada ödem gözlendi. Kateşin + klorprifos ve kuersetin + klorprifos uygulanan gruplarda mikroskobik bulguların hafif şiddette olduğu gözlendi. Sonuç olarak kateşin ve kuersetinin, klorprifosun meydana getirdiği hasarı azalttığı tespit edildi.

Chlorpyrifos induced small intestine toxicity in rats and protective role of quersetin and catechin

Abstract

In this study we investigated that chlorpyrifos which is a widely used an organophosphorous pesticide induced small intestine toxicity and protective role of quersetin and catechin. For this reason, chlorpyrifos (5.4 mg/kg 1/25 LD50), catechin (20 mg/kg), quercetin (20 mg/kg), catechin (20 mg/kg) + klorprifos (5.4 mg/kg 1/25 LD50) and quercetin (20 mg/kg) + chlorpyrifos (5.4 mg/kg 1/25 LD50) were given to male rats orally for four weeks. At the end of fourth week, histopathological changes in the small intestine tissues were investigated using light microscope comparatively with control group. At the end of the 4th week, there were no histopathological changes between the catechin-treated and quercetin-treated groups compared to the control group. After four weeks of chlorpyrifos exposure shortening and necrosis in intestinal villi and edema in muscular mucosa were observed in the small intestine tissues. Milder histopathological changes were observed in animals co-treated with catechin plus chlorpyrifos or quercetin plus chlorpyrifos. As a result, it appears that catechin and quercetin moderated chlorpyrifos-induced toxicity.
