Effects of Some Applications on Anthocyanin Production in Callus Culture of Black Carrot (Daucus carota ssp. sativus var. atrorubens Alef)
Abstract
In this study, effect of some applications on the production of
anthocyanin in the callus culture of black carrot (Daucus carota ssp. sativus var.
atrorubens Alef.) were studied. Hypocotyle explants excised from sterile seeding
were cultured on MS (Murashigeand ve Skoog, 1962) media containing 2 mg/L
NAA (Napthalene acetic acid), 0,2 mg/L BAP (Benzylaminopurine), 30 g/L
sucrose and 2 g/L phytagel. Riboflavin and UV-C as an elicitor were applied to
callus culture at different concentrations and durations in order to increase the
production of anthocyanin. Callus tissues were exposed to UV-C (254 nm) light
at 10 cm distance from the source for 10 and 15 minute in a day. After UV-C
application, callus tissues were incubated under 16 h. light and 8 h. dark
photoperiod at 25 0C ± 2 and harvested after 10 days. The effects of riboflavin at
the concentrations of 4, 8 and 10 mg /L on enhancement of anthocyanin
production were studied. In a callus culture of black carrot, anthocyanin
biosynthesis was strongly enhanced, whereas cell growth was inhibited, eliciting
either the addition of riboflavin or UV-C light irradiation. The highest values of
anthocyanin production were obtained from callus cultures applied 4 mg/L
riboflavin (1.18 mg/g FCW) and 15 min. UV-C (1,02 mg/g FCW), attaining
amounts 2,51 and 2,17 times greater than the control, respectively
