Abstract

HPLC and its Recent Applications to Foods Chromatography is a separation technique; By means of this technique, compounds in the solution can be separated individually as a result of their different movement by influence of moving phase on the porous stationary phase. There are many kinds of chromatography, but chromatographic methods, generally, falls into two main groups, depending on the state of moving phase, when moving phase is gas or liquid, chromatography is called gas (GC), or liquid chromatography (LC), respectively. High performance liquid chromatography (HPLC) is included in LC. Since liquid carrying the sample is eluted through column under pressure obtained by pneumatic or mechanic pump device, a more precise and quicker separation is carried out than usual chromatography. It is obvious that HPLC has great advantage in the analysis of heat-sensitive constituents of foods, because it is not necessary to vaporize the sample.

Abstract

Bir karışım veya çözeltide bulunan ayrı ayrı maddelerin durgun gözenekli bir ortamda, hareketli bir çözücünün etkisiyle farklı hızlarda hareket etmeleri sonucu birbirinden ayrılmaları işlemine kromatografi denir. Birçok çeşidi olmakla beraber, kromatografi, genelde hareketli fazın gaz veya katı durumda oluşuna göre gaz kromatografisi (GC) veya sıvı kromatografisi (LC) diye iki ana bölüme ayrılabilir. Yüksek performanslı sıvı kromatografisi (HPLC) sıvı kromatografisi içinde yer almaktadır. Bu kromatografide örneği taşıyıcı sıvı, pnömatik veya mekanik pompalarla basınç altında kolondan geçirildiğinden, sıvı fazın kendi halinde kolondan geçişine göre çok daha yüksek hareketli faz hızı sağlanmakta ve ayrılma çabuk ve tam olmaktadır. Isıya duyarlı ve gaz haline getirilmeleri mümkün olmayan gıda bileşenlerinin analizinde HPLC’nin özel bir önemi vardır.
