Analysis of High Frequency Conjugal Transfer Systems in Strains of Lactococcus lactis Subsp. Lactis (Turkish with English Abstract)

Abstract

In this study, high frequency conjugal transfer ability of six different lactose plasmids encoding lactose fermentation in L. lactis subsp. lactis strains was searched. Depending on the interactions between the two sex factors (Clu, and Agg), the conjugal transfer frequencies of these plasmids have ranged in the rates of 1.5x10-5-1.0x10-7 for Clu-/Agg-, Agg+xClu-/Agg+, Agg- or Clu+/Agg-xClu-/Agg- conjugation pairs and 7.1x10-2-2.7x10-3 for Clu+/Agg-xClu-/Agg+ conjugation pairs. And the stability of the lactose plasmids were 82-96% in wild type strains, 77-98% in the conjugants of recipient strain MG1390, 44-67% in the conjugants of recipient strain MCL8060.

Lactococcus lactis Subsp. Lactis Suşlarında Yüksek Sıklıkta Konjugal Transfer Sistemlerinin Analizi

Abstract

Bu çalışmada L. lactis subsp. lactis suşlarında laktoz fermentasyonu özelliğini kodlayan altı farklı plazmidin yüksek sıklıkta konjugal aktarım yeteneği araştırıldı. Bu plazmidlerin konjugal transfer sıklıkları; iki seks faktörünün interaksiyonuna bağlı olarak (Clu ve Agg), Clu-/Agg-, Agg+ x Clu-/Agg+, Agg- ya da Clu+/Agg- x Clu-/Agg- konjugasyon eşleri için 1.5x10-5–1.0x10-7 ve Clu+/Agg- x Clu-/Agg+ konjugasyon eşleri için 7.1x10-2-2.7x10-3 oranlarında değişim gösterdi. Laktoz plazmidlerinin stabiliteleri ise; doğal suşlarda %82-96, MG1390 alıcı suşu için tanımlanan konjugantlarda %77-98 ve MCL8060 alıcı suşu için tanımlanan konjugantlarda ise %44-67 arasında saptandı.
