HIZLI Escherichia coli TANIMLANMASINDA ULTRAVİYOLE DESTEKLİ İNDOL VE MUG ESASLI TESTLERİN KIYASLANMASI

Abstract

Bu çalışmada önceki ISO standardında (ISO 9308-1:2000) belirtilen UV destekli indol esaslı hızlı E. colitesti ile Bactident E. coli testi (MUG esaslı hızlı test) karşılaştırılmıştır. Bu amaçla çeşitli gıda örneklerindenizole edilen ve tanısı yapılmış olan 46 adet E. coli izolatı, 4 referans E. coli suşu ile koliform grup bakterileringeliştirildiği besiyerlerinde tipik koliform kolonisi oluşturmasa da; gelişerek koloni oluşturabilen bireradet Salmonella Enteritidis ve Klebsiella oxytoca olmak üzere toplam 52 Enterobacteriaceae üyesibakteri her iki yöntemle analiz edilmiştir. Araştırma sonuçlarına göre MUG esaslı hızlı test %100 doğrusonuç verirken UV destekli indol esaslı testte %94.1 doğru sonuç alınmıştır. K. oxytoca, bu testte sorunaneden olmuştur. Laboratuvar koşularında Bactident E. coli kitinin kullanılması UV destekli indol testinekıyasla çok daha hızlı ve kolay olarak değerlendirilmiştir

THE COMPARISON OF RAPID Escherichia coli TESTS BASED ON MUG AND ULTRAVIOLET SUPPORTED INDOLE

Abstract

In this study, Bactident E. coli (rapid test based on MUG) and indole test supported by UV which wasindicated in previous ISO method (ISO 9308:2000) was compared. Both methods were applied on 46E. coli strains which were isolated from different food samples, 4 reference E. coli strains, 2 non-coliformbacteria as Salmonella Enteritidis and Klebsiella oxytoca which can be grown on the coliform media.46 E. coli isolates, isolated from various foods and identified by MUG and indole tests. According toresults accuracy of indole test supported by UV was determined as 94.1% whereas it was found 100%for Bactident E. coli. K. oxytoca has caused problem in UV supported indole test. In laboratory conditions,using Bactident E. coli is more rapid and easy then UV supported indole test
