Abstract

ABSTRACT

Spider venoms are biologically active substances which affect a variety of vital physiological functions in both insects and mammals. The major constituents of spider venoms are protein, polypeptide and polyamine neurotoxins, enzymes, nucleic acids, free amino acids, monoamines and inorganic salts. The identification of the proteins in spider venoms is an essential step for identification of venom. In this study, we compared with protein profiles of Agelena labyrinthica venom obtained by electrostimulation of the prosoma and extracted directly from gland. There were seven components identified in whole venom, when whole venom and venom gland extracts composition of A. labyrinthica were compared by SDS-PAGE.

Abstract

Örümcek zehirleri; hem avları olan böceklerde hem de memelilerde hayatsal fizyolojik fonksiyonları etkileyen biyo-aktif maddeler olup esas bileşenleri; protein, polipeptid, poliamin nörotoksinler, enzimler, nükleik asitler, serbest amino asitler, monoaminler ve inorganik tuzlardır. Örümcek zehirinde bulunan proteinlerin tanımlanması, zehirin tanımlanmasında önemli bir adım teşkil etmektedir. Bu çalışmada; ülkemizde oldukça yaygın bir örümcek türü olan Agelena labyrinthica’nın uyarılmamış ve uyarılmış durumdaki zehir bezlerinden elde edilen protein profilleri saf zehir salgısı ile karşılaştırılmıştır. Uyarılmamış zehir bezi ekstraktı ve elektriksel uyarımla elde edilmiş saf zehirin protein profilleri, SDS-PAGE analitik yöntemi kullanılarak karşılaştırıldığında, zehir proteininin yapısal proteinlerden kesin olarak ayrıldığı ve saf zehirde molekül ağırlıkları bakımından yedi farklı polipeptidin bulunduğu belirlenmiştir
