Bir Siyah Kuğuda (Cygnus atratus) PZR ile Cinsiyet Tespiti

Abstract

Bu çalışmada, Harran Üniversitesi Veteriner Fakültesi’ne getirilen bir siyah kuğunun (Cygnus atratus) cinsiyetinin polimeraz zincir rekasiyonu (PZR) yöntemi ile belirlenmesi amaçlanmıştır. Bu amaçla CHD (chromodomain-helicase-DNAbinding) lokusunun hedef bölgesi iki farklı primer çifti (1237L-1272H ve P2-P8) yardımıyla PZR yöntemi ile çoğaltıldı ve PZR ürünleri agaroz jel elektorofrezi ile ayrımlandı. Her iki primer çifti ile elde edilen PZR ürünleri için agaroz jelde farklı uzunlukta iki bant gözlendi. Kuğunun dişi olduğu kanaatine varıldı.
