Identification and Phage Susceptibilities of Brusella Strains In Turkey Between the Years 2007 and 2008

Abstract

A total of 114 Brucella isolates consisting of 43 cattle, 61 sheep and goats, 8 human and 2 food origins were identified by conventional biotyping procedures. The results showed that 96% of cattle isolates was B. abortus biotype 3 and 4% was B.melitensis. One of B. melitensis was B.melitensis biotype 3 and the other was B. melitensis biotype 1. Of sheep and goat isolates, 65.6 % was B. melitensis biotype 3 and 29.5% was B. melitensis biotype 1. Three of the sheep strains (4.9 %) were identified as B. abortus biotype 3. None of the test strains was identified as B.abortus S19 ve B.melitensis Rev.1 vaccine strains. Seven out of 8 strains from humans were identified as B. melitensis biotype 3. One strain of human origin was lysed completely by R/C phage. This isolate was not agglutinated by anti M and anti A monospecific sera and was agglutinated by both R antisera and acriflavin displaying rough characteristics. Two of the strains from food (ice cream) were B. melitensis biotype 1 and B. melitensis biotype 3. Four of B. melitensis strains were are atypically susceptible to penicilin and 2 of the B. melitensis strains were evaluated as atypical regarding to their susceptibility to basic fuhsin (20 µg/ml). All the B.melitensis strains were tested for the susceptibility to the phages of Tbilisi, Berkeley, Weybridge, Izatnagar and R/C at routine test dilutions (RTD). Forty seven (67.2%), 15 (21.4%) and 8 (11.4%) out of 70 B. melitensis isolates conformed to phage typing pattern C, D and B, respectively.

Türkiye’de 2007 ve 2008 Yılları Arasında İzole Edilen Brusella Suşlarının İdentifikasyonu ve Faj Duyarlılıklarının Saptanması

Abstract

Çalışmada 43 sığır, 61 koyun, 8 insan ve 2 adet gıda orijinli olmak üzere toplam 114 brusella suşu konvensiyonel biyotiplendirme prosedürü ile biyotiplendirildi. Sığır kökenli suşların % 96’sı B.abortus biyotip 3 ve % 4’ü B.melitensis olarak identifiye edildi. B.melitensis suşlarının biri B.melitensis biyotip 3 ve diğeri B.melitensis biyotip 1 idi. Koyun ve keçi kökenli suşların % 65.6’sı B.melitensis biyotip 3 ve % 29.5’u B.melitensis biyotip 1 olarak identifiye edildi. Koyun suşlarından 3’ü (%4.9) ise B.abortus biyotip 3 olarak tespit edildi. Test suşlarından hiçbiri B.abortus S19 ve B.melitensis Rev.1 aşı suşları olarak saptanmadı. İnsanlardan izole edilen 8 suşun 7’si B.melitensis biyotip 3 olarak ve dondurmadan izole edilen suşlardan biri B.melitensis biyotip 3 ve diğeri B.melitensis biyotip 1 olarak idendifiye edildiler. İnsandan izole edilen suşlardan birinin rough özellikte ve R/C fajı ile tamamen lize olduğu tespit edildi. B.melitensis suşlarından 4’ü atipik olarak penisiline duyarlılık gösterirken, 2’si bazik fuksine (20 µg/ml) gösterdiği duyarlılık nedeni ile atipik olarak değerlendirildi. Tüm B.melitensis suşları rutin test dilüsyonundaki (RTD) Tbilisi, Berkeley, Weybridge, Izatnagar ve R/C fajlarına duyarlılık açısından test edildiler. Toplam 70 B.melitensis suşunun 47’si (%67.2) C, 15’i (%21.4) D ve 8’i (%11.4) B faj tip kalıbına uyan reaksiyonlar gösterdi.
