Effects of various osmotic pressure levels and cryoprodective agents (CAPs) on Awassi Ram Spermatozoa

Abstract

Effective ram sperm cryopreservation protocols, which would yield acceptable fertility rates following artificial insemination (AI) in ewes, are currently lacking. During the process of cryopreservation, spermatozoa are exposed to osmotic pressure and heat stress at different rates. To reduce these negative effects on spermatozoa, new freezing methods and extenders are being tried to develop. The objectives of the current studies are to compare the effects of various anisoosmatic conditions, cryoprotective agents (CAPs) on the motility, acrosome and membrane integrity of Awassi sperm. In the mating season, the sperm was collected from 2 Awassi ram than has been mixed and used. Two experiments were conducted. In experiment 1, the sperm was exposed to 75, 150, 225, 325, 425, 600 and 900±5 mOsm/kg sucrose solutions, held for 5 min and then returned to isosmotic condition. In experiment 2, the sperm was exposed to 0.5, 1.0 and 1.5 M glycerol (Gly), dimethyl sulfoxide (DMSO), ethylene glycol (EG) and propylene glycol (PG) for 5 min and then returned to isosmotic condition. The sperm was evaluated by microscopic assessment of the sperm motility, acrosome and membrane integrity. The motility of ram sperm was significantly affected from anisosmotic stress (P< 0.05). While anisosmotic stress had no effects on acrosomal integrity of ram sperm, there was a significant reduction in acrosomal integrity for ram sperm after the addition and removal of a 75 mOsm sucrose solution. The abrupt addition and removal of cryoprotectant had effect on the motility and acrosomal integrity of epididymal ram sperm (P < 0.05). There was a decrease in acrosomal integrity for ram sperm after exposure to cryoprotectant (P 0.05). In conclusion, the current data suggest that ram sperm was affected by cryobiologiably stress conditions at different rates.
