Investigation of Usefulness of DNA Microsatellite Markers in Some Carp Species
Abstract
In this study; application of different microsatellite markers were investigated for three species (Capoeta trutta
(Heckel, 1843), Barbus luteus (Heckel, 1843) ve Cyprinion macrostomus (Heckel, 1843)) belonging to Cyprinidae family.
Taking muscle tissue from the fishes that are caught from the Euphrates and Tigris rivers system, DNA extraction was
performed by using kits. Then, the usefullness of Polymerase Chain Reaction (PCR) system was searched by using eight
microsatellite markers (MFW-1, CyPG3, CyPG8, SarN7G5, Barbus27, Barbus31, Barbus33, Barbus50). The PCR products were
then obtained in four of them (MFW-1 CyPG3, Barbus27, Barbus3) by amplification. The fragment analysis of the resulting
products were performed by capillary electrophoresis in order to identify te alleles of individuals. As a result, it is possible to
benefit from these four microsatellite markers in genetic diversity studies.
