The Effect of Enrofloxacin on Myometrial Contractility in Bitches
Abstract
Enrofloxacin is a broad spectrum antibiotic used to treat bacterial infections. The aim of present study was to
investigate the effect of enrofloxacin on Prostaglandin F2 alpha (PGF2 alpha) induced postpartum canine myometrium in
vitro. Uterine tissues were obtained from bitches undergoing ovariohysterectomy in the postpartum period. Isolated
myometrium strips suspended in organ bath filled with Kumagai solution and continuously gassed with 95% oxygen and 5%
carbon dioxide (28°C). At the end of equilibration period, PGF2 alpha was added to the organ bath and the myometrial
strips were observed to be contracted. Then the organ bath were administered solvent (n = 6), 2 mM (n = 6) and 5 mM (n =
10) enrofloxacin and % contraction was monitored. PGF2 alpha was given a second time and the contractions were
recorded after incubation with enrofloxacin. Solvent inhibited significantly myometrial contractions and enrofloxacin
treated group showed a biphasic effect. The contractile response was significantly inhibited by 5 mM enrofloxacin
compared to the control group. Our results show that high-dose enrofloxacin had inhibitory effects on the canine
myometrial contractions. For this reason, enrofloxacin should be used combine with uterotonic agents in the metritis
treatment.
