Immunohistochemical Investigation in Sheep with Parasitary Liver Fibrosis

Abstract

Liver fibrosis significantly effect human and animal health in our country and worldwide. In this study, 30 sheep liver tissues with macroscopical parasitary fibrosis and 10 healthy liver tissues were taken in terms of histopathologically and immunohistochemically (α-SMA, iNOS COX-2, p53 and PCNA). Cell infiltration consisting of lymphocytes, macrophages and eosinophils and fibrosis were detected in portal area in histopatological investigation. And also there was nodules (pseudolobulus) formed by regenerated hepatocytes that do not vena centralis in center. Also granulomas contained in foreign body giant cells were seen. According to immunohistochemical examination; stronger positivity was observed in α-SMA, iNOS, COX-2, p53 and PCNA compared to control group. It has been found beneficial to use α-SMA to demonstrate connective tissue uptake in fibrosis, using COX-2 as due to fibrosis and cirrhosis at the base of liver tumors and their role in wound healing; using PCNA in order to evaluate increase proliferation, using p53 for apoptosis, using iNOS antibody as its secreted from macrophages in parasitic infections.

Koyunlarda Paraziter Kaynaklı Karaciğer Fibrozisinde İmmunohistokimyasal İncelemeler

Abstract

Karaciğer fibrozisi, ülkemizde ve dünyanın pekçok bölgesinde insan ve hayvan sağlığını önemli ölçüde etkilemektedir. Bu çalışmada, makroskobik incelemede paraziter fibrozis bulguları gösteren 30 koyun karaciğeri histopatolojik ve immunohistokimyasal olarak (α-SMA, iNOS COX-2, p53 ve PCNA) incelendi. Histopatolojik incelemede portal alanlarda lenfosit, makrofaj ve eozinofilden oluşan hücre infiltrasyonu ile fibrozis, parankimde vena sentralisleri bulunmayan rejenere hepatositlerin oluşturduğu nodüller (pseudolobulus) şekillenmişti. Ayrıca bazı olgularda içinde yabancı cisim dev hücreleri bulunan granülomlar görüldü. İmmunohistokimyasal incelemede; kontrol grubuyla karşılaştırıldığında fibrotik karaciğerlerde α-SMA, iNOS, COX-2, p53 ve PCNA için çok daha yüksek pozitiflik gözlendi. Fibroziste bağ doku artışını göstermek için α-SMA, karaciğer tümörlerinin temelinde fibrozis ve sirozun olması ve yara iyileşmesinde rol alması yönünden COX-2, artan proliferasyonun değerlendirilmesinde PCNA, apoptozis için p53; paraziter enfeksiyonlarda makrofajlarda salınması yönünden iNOS antikorunun kullanılmasının faydalı olduğu görülmüştür.
