Lilium candidum L.’da In vitro Mikroçoğaltım ve Alkaloidler

Abstract

Bu çalışmada in vitro koşullarda mikroçoğaltım yöntemleriyle üretilen soğanlarda gelişimin gözlendiği dönemden itibaren 3 farklı dönemde (1., 2. ve 3. ay sonunda) soğancıklardan örnek alımı yapılmıştır. Örnek alınan soğancıkların ekstraksiyonu yapıldıktan sonra alkaloid (azotlu bileşik) profili GC-MS ile tanımlanmıştır ve in vivo’da yetişen soğanlarla karşılaştırarak mikroçoğaltım çalışmalarında in vivo koşullara en yakın örnek alım dönemi belirlenmiştir. Çalışmada L. candidum’un soğan pul yaprakları eksplant olarak kullanılmış ve rejenerasyon sonucu 3 farklı besin ortamından ve 3 farklı dönemde alınan soğancıklarda GC-MS analizi yapılmıştır.  Yapılan GC-MS analizi sonucunda toplam alkaloid yüzdesi kontrole (%13.11) en yakın örnek 2. besin ortamının (0.2 mg/L NAA + 0.4 mg/L TDZ) 3. örnek alımından (%7.42) elde edilmiştir. Belirlenen en yüksek yüzde alana sahip alkaloid (1H-Pyrrole, 2,5-dihydro-1-nitroso-) ise yine 2. besin ortamının 3. örnek alımında belirlenmiştir.
