Metallocenes and Their Use in Health

Abstract

There are some structural and chemical similarities between metallocene dihalide [M(η5-Cp)2X2] and the cis-diaminedichloroplatin(II) complex, which is very well known as a tumor agent. Studies have shown that the biochemical behavior of metallocenes is different and it has been determined that it has an antitumor effect incidentally. It has been observed that metallocene dihalide compounds have high activity against tumor cells. The most important evidence for this is metal ion accumulation in tumor cells. As a result of biochemical studies, it has been determined that the activity of metallocene complexes is strongly affected by the central ion. It was observed that the separation of cyclopentadienyl (Cp) ligand from the structure varies depending on the metal ion and the pH of the solution. For example, while it was detected that the cyclopentadienyl ring was separated from the structure in the Ti4+ complex, this was not observed in the V4+ complex. Accordingly, it has been determined that the biological activities of these complexes differ. In this review, information is given on the chemical structure of metallocenes, the synthesis of metallocene complexes and their use in medicine in cancer treatment.

Abstract

Metalosen dihalojenür [M(η5-Cp)2X2] ile çok iyi tümör ajanı olarak bilinen cis-diamindikloroplatin(II) kompleksi arasında bazı yapı ve kimyasal benzerlikler vardır. Yapılan çalışmalar metalosenlerin biyokimyasal davranışlarının farklı olduğunu ortaya koymuş ve tesadüfi olarak antitümör etkisi gösterdiği belirlenmiştir. Metolasen dihalojenür bileşiklerinin, tümör hücrelerine karşı yüksek aktiviteye sahip olduğu gözlenmiştir. Bunun en önemli kanıtı olarak, tümör hücrelerindeki metal iyonu birikimi gösterilmiştir. Biyokimyasal araştırmalar sonucunda metalosen komplekslerinin aktivitesinin merkez iyonundan güçlü bir şekilde etkilendiği tespit edilmiştir. Siklopentadienil (Cp) ligandının yapıdan ayrılmasının metal iyonuna ve çözeltinin pH’sına bağlı olarak değiştiği gözlenmiştir. Örneğin, Ti4+ kompleksinde siklopentadienil halkasının yapıdan ayrıldığı tespit edilirken, V4+ kompleksinde bu durum gözlenmemiştir. Buna bağlı olarak da bu komplekslerin biyolojik aktivitelerinin farklılık gösterdiği belirlenmiştir. Bu derlemede metalosenlerin kimyasal yapısı, metalosen komplekslerinin sentezlenmesi ve tıpta kanser tedavisinde kullanımı üzerine bilgi verilmektedir.
