Teke Spermasının Kriyokonservasyonu ve Uygun Yöntem Kullanımı

Abstract

Kriyokonservasyon, üreme biyoteknolojisi, ıslah ve genetik kaynakların korunma ve muhafazası gibi birçok alanda önemli fayda sağlamaktadır. Uygun kriyokonservasyon yöntemleri sayesinde üremede başarı arttırılabilecektir. Özellikle keçiler gibi mevsime bağlı üreme yapısına sahip türlerde kriyokonservasyon büyük avantaj sağlamaktadır. Türlere göre üreme biyoteknolojilerinin geliştirilmesi, sahip oldukları biyolojik farklılıkların bir sonucu olup, dondurma çözdürme sonrası hayatta kalma oranı türlere göre önemli farklılık göstermektedir. Sperma membranının lipid ve protein gibi kompozisyonunu oluşturan bileşenlerin çözdürme sonrası kalite üzerinde etkili olduğu bilinmektedir. Teke spermasının özelliklerinin belirlenerek uygun semen alımı, değerlendirilmesi işlemleri ile dondurma protokollerinin geliştirilmesi üreme performansını arttırıcı önemli unsurlar olup, yapay tohumlama ya da diğer in vitro – in vivo çalışmalarda spermin kullanılabilirliğini etkilemektedir. Kriyokonservasyonun zararlı ve öldürücü etkisinin azaltılması dondurma çözdürme sonrası sperm kalitesi üzerinde doğrudan etkilidir. Dondurmada kullanılan çözelti ve biyolojik ortamların iyileştirilmesi gerekmektedir. Bu bağlamda tekelerde semen alımı, işlenmesi, kriyokonservasyonu ve çözdürme süreçlerini kapsayan protokollerin maliyet, iş gücü ve zaman tasarrufu da sağlayarak uygun sperm analiz yöntemlerin oluşturulması gerekmektedir.
