Abstract

Özet: Stres vücudun çevreden gelen taleplere karşı adaptasyonunu sağlayan ve hayatta kalabilmek için gerekli olan bir yanıtıdır. Ancak yaşamın erken evrelerinde maruz kalınan stres, erişkin bireyin büyüme ve gelişimi üzerinde derin etkilere sebep olabilir. Yapılan çalışmalar, gebelik sırasında veya erken yaşam dönemlerindeki stresli deneyimlerin, ruhsal ve zihinsel bozuklukların oluşmasına zemin hazırladığını göstermektedir. Gebelik sürecinde maruz kalınan stres, birkaç farklı mekanizma aracılığıyla fetüsün beyin gelişimi ve davranışları üzerinde olumsuz etkilere neden olmaktadır. Bu mekanizmalardan ilki; stresin hipotalamo-pituiter-adrenokortikal (HPA) aksın negatif geribildirim regülasyonunda oluşturduğu bozukluklar ve glukokortikoid düzeylerindeki artıştır. İkincisi; GABAerjik, serotoninerjik ve dopaminerjik yolakların aktivitesindeki değişikliklerdir. Üçüncü mekanizma ise; sempatoadrenal sistemdeki aktivite artışına sekonder oluşan değişiklikleri içermektedir. Tüm bu olumsuz etkiler sonuçta fetüsün stres hassasiyetini arttırmakta ve bireysel yatkınlık düzeyleri ile de ilişkili olarak, uzun vadede psikopatolojik süreçlerin gelişimine neden olmaktadır. Stres maruziyetine bağlı olarak meydana gelen fonksiyonel ve anatomik değişikliklerin açığa çıkarılması amacıyla deney hayvanları üzerinde yapılan çalışmalar; stresörlerin dozuna, süresine, şiddetine ve zamanlamasına müdahale etmeye imkân vermesi bakımından önemli avantajlar sunmaktadır. Bu derlemede, erken yaşam dönemlerindeki stres maruziyetinin yol açtığı değişikliklere ilişkin deneysel çalışmalardan elde edilen verilerin gözden geçirilmesi amaçlanmıştır. Bu değişikliklere yol açan molekül ve mekanizmaların aydınlatılması, koruyucu veya daha etkili tedavi stratejilerinin geliştirilmesi açısından yararlı olacaktır.

NEUROBIOLOGICAL EFFECTS OF PRENATAL STRESS EXPOSURE

Abstract: Stress is an adaptive response against environmental demands and it is required to survive. However, exposure to early life stress might cause profound effects on the growth and development of an adult individual. Recent studies show that stressful experiences during pregnancy or early life stages might increase the risk of emotional and mental disorders. Exposure to stress during pregnancy causes adverse effects on the development of brain and behaviours of the fetus via several different mechanisms.  The first mechanism is the disturbed regulation of hypothalamic-pituitary-adrenocortical (HPA) axis negative feedback and enhanced glucocorticoid levels. Second one is the alternations in the activity of GABAergic, serotonergic and dopaminergic pathways. The third mechanism consists of the changes occurring secondary to enchanced activity of sympathoadrenergic system. All of these negative effects then increase the fetal sensitivity to stress and, in the long term, cause development of psychopathological processes associated with the susceptibility level of an individual. Animal studies that reveal the functional and anatomical changes occurring due to the stress exposure, offer significant advantages since they allow altering the dose, duration, severity and timing of the stressors. In this review, we aimed to evaluate the data obtained from experimental studies on the early life stress exposure. Elucidation of the molecules and mechanisms causing this process might be beneficial for the development of more effective prevention or treatment strategies.
