Bakır Asetoarsenit'in İn Vivo Genotoksik Etkilerinin Değerlendirilmesi

Abstract

Mevcut çalışmada, Sprague-Dawley sıçanlarının kemik iliği hücreleri üzerinde Bakır asetoarsenit (BAA) in genotoksik etkileri araştırıldı. Bu amaçla, sıçanlar rastgele altı gruba ayrıldı ve her bir grupta yedi hayvan yer aldı. Kontrol grubu hariç, BAA dozları (5, 10, 15, 25 ve 30 mgkg-1) deney hayvanlarına beş gün boyunca intraperitonal (i.p) yolla verildi. BAA enjeksiyonlarından sonra sıçanlar anestezi edildi ve bu sürenin sonunda kemik iliği hücreleri alındı. Kromozom aberasyonlarını (KA) tespit etmek için preparatlar Giemsa ile boyandı ve ışık mikroskobu ile incelendi. Bunun yanı sıra, mikroçekirdek (MÇ) frekansı May-Grünwald ve Giemsa ile boyanmış preparatlarda değerlendirildi. Kontrol grubuyla karşılaştırıldığında, BAA ile muameleden sonra yapısal KA sayısı ve MÇ frekansları belirgin bir şekilde arttı. Aynı zamanda, kemik iliği hücrelerinde BAA in genotoksik etkilerinin doza bağlı olarak arttığı tespit edildi.
