Investigation of Potential Drug-Drug Interactions Due to Linezolid Use in Anesthesia Intensive Care Unit: A Retrospective Study

Abstract

Objective This study aims to clarify the possible interactions of Linezolid, a drug used during infection treatment in intensive care units, with other drugs and to provide information to stakeholders in clinical use. Material and Method Data from the files of intensive care patients over the age of 18 who received at least 24 hours of inpatient treatment between 2017-2023 in the intensive care unit of the Department of Anesthesiology and Re- animation in Suleyman Demirel University and who used at least one other drug in addition to the active substance Linezolid during this period were used. All these data were evaluated with the Lexi-Interact pro- gram. Results 68(36%) patients included in the study were exposed to polypharmacy for 2-22 days, 69(36.5%) patients for 23-45 days, and 52(27.5%) patients for 46-197 days during their treatment. A total of 655 interactions were identified. Of these, 17(8.99%) type A, 172(91%) type B, 183(96.83%) type C, 184(97.35%) type D, and 99(52.38%) type X interactions were detected. As a result of the examination of potential drug-drug inter- actions (pDDIs) in terms of dual drug use with Linezol- id, it was seen that the most frequent type C interac- tion could occur with Quetiapine (33;17.20%), type D interaction with Salbutamol (94;49.70%), and type X interactions which are not recommended for use side by side with Linezolid could occur with Metoclopropa- mide (44;23.30%) and Escitalopram (11;5.80%). Conclusion Linezolid-treated ICU patients exhibited a high preva- lence of potential drug–drug interactions, particularly involving sympathomimetic and serotonergic agents. These findings highlight the importance of routine interaction screening and careful monitoring when linezolid is prescribed in critically ill patients.

Our study was conducted retrospectively after receiving approval from the ethics committee of Suleyman Demirel University Faculty of Medicine (dated 16/08/2023-no.13/184) by examining the files of patients over the age of 18 who were treated as inpatients for at least 24 hours between 2017 and 2023 in the iıntensive care unit of the Department of Anesthesiology and Reanimation of Suleyman Demirel Faculty of Medicine and who used at least one different medication in addition to the active substance called Linezolid during this period.
