Akut diazinon toksisitesinin testis dokusunda oluşturduğu histolojik değişiklikler ve bu değişikliklere C vitamini ve E vitaminin etkisi

Özet

Amaç: Akut diazinon toksisitesinde sıçan testis dokusunda oluşan yapısal değişiklikler ve bu değişikliklere E ve C vitaminlerinin etkilerinin araştırılması amaçlandı. Materyal ve Metod: 200-290 gram (gr) ağırlığında, 24 adet Wistar albino türü erkek sıçan kullanıldı. Grup I'e (kontrol grubu) oral olarak mısır yağı verildi. Grup II'ye mısır yağında eritilmiş diazinon (Bazidin) preparatı 335mg/kg dozunda oral olarak, Grup III'e 335 mg/kg dozunda diazinon oral olarak, 200 mg/kg C vitamini (Redoxan) intraperitoneal olarak, 150 mg/kg E vitamini (Evigen) intramüsküler olarak uygulandı. Histolojik inceleme için rutin takip yöntemleri uygulandıktan sonra elde edilen testis dokusu kesitlerinde her grup için ayrı ayrı Johnsen testiküler biyopsi skoru hesaplandı ve histolojik değerlendirme yapıldı. Bulgular: Johnsen testiküler biyopsi skoru değerlendirmesinde Grup II ve III, Grup I ile karşılaştırıldığında skordaki azalış istatistiksel olarak anlamlı idi (p‹0.05). Grup II ve Grup III arasındaki biyopsi skoru farkı ise istatistiksel olarak anlamlı değildi. Grup II ve III'de germinal epitel diziliminde organizasyon bozukluğu tespit edildi. Sonuç: Akut tek doz diazinonun testis dokusu üzerine toksik etkileri olduğunu saptadık. Fakat bu yüksek doz diazinonun etkisi ile oluşan morfolojik değişikliklerin belirtilen doz ve sürede verilen E ve C vitamini kombinasyonu ile önlenemediğini tespit ettik.

Abstract

Histological changes in acute diazinon toxicity in testis tissue and effects of Vitamin C and Vitamin E on structural changes


Aim: It was aimed to investigate the effects of a single dose diazinon on adult rat testis tissue and the protective effects of vitamin E and C on damage. Materials and Methods: 24 male Wistar albino rats, weighing 230-280 gr were divided into three groups. Group I were given corn oil, Group II were given 335 mg/kg diazinon (Bazidin) solved in corn oil, Group III were given 335 mg/kg diazinon, 200 mg/kg vitamin C (Redoxan), 150 mg/kg vitamin E (Evigen). Histologic evaluation of testis tissue including Johnsen testiculer biopsy score was performed for each group after practicing routine histologic procedures. Results: Johnsens testiculer biopsy score of Group II and III were significantly decreased when compared to Group I (p<0.05). Difference between Group II and III was not significant. In Group II and III germinal epitel disorganization was observed. Conclusion: We have observed that single high dose of diazinon has toxic effects on rat testis tissue. This morfological alterations were not healed by the vitamin E and C administration at this dose.
