Plant Regeneration from Mature Embryo of Orchardgrass (Dactylis glomerata L.)

Abstract

In this research, response of mature embryos of two wild ecotypes and two cultivar of orchardgrass (Dactylis glomerata L.) to tissue culture were investigated. In the study, 2 auxin types (dicamba and picloram), 3 different doses (1,5 mg/l, 2,0 mg/l and 2,5 mg/l) and 2 different doses of sucrose at callus stage were used. Although the effect of applications were not significant on callus formation ratio, it was significant on plant regeneration. Plant regeneration varied according to genotypes, hormone types and hormone doses. According to the results; 1,5 mg/l dose of picloram was found appropriate for plant regeneration from mature embryo.
