The Identification of Turkish Honey Bee Populations Inferred From Mitochondrial DNA Cytochrome C Oxidase I and Cytochrome C Oxidase II Intergenic Region (COI-COII Intergenic Region)
Abstract
In this study, the identification of Turkish honey bee populations was carried out using cytochrome C oxidase I and II intergenic region of mitochondrial genome. A total of 244 worker bees from 20 different locations in Turkey were used. Restriction fragment length polymorphism (RFLP) and DNA sequence data were utilized to identify the honeybee populations. DraI digestion of the COI-COII intergenic region generated four fragments. In this study, DraI digestion of the COI-COII intergenic region gave rise to a total of five DraI haplotypes which have been accurately identified from DNA sequence results. Three of the DraI haplotypes (47/41/64/420 bp-C1; 47/40/64/420 bp-C2 and 47/39/64/420 bp) have been reported in other papers but two of them (47/41/64/419 bp and 47/39/64/418 bp) were first seen in this study. DraI digestion of the COI-COII intergenic region generated four fragments in Turkish honeybees which previously have been found to be characteristic of the C lineage an these results confirmed that Turkish honey bee populations belong to East European and Mediterranean (C) lineage.
