Determination of Genetic Diversity Among Chickpea (Cicer arietinum L. ) Populations Collected from The Konya Region Using ISSR Markers

Abstract

This study was conducted to determine the genetic diversity of the chickpea populations collected from different localities in Konya, an important region in chickpea production of Turkey. Support of breeding programs that target the development of chickpea varieties with economic value that are also appropriate to the region were also aimed. In the present study, 23 chickpea populations collected from the Konya region were used together with two commercial varieties (Gökçe, Er 99) and genetic distance of the populations were determined via inter simple sequence repeats (ISSR) molecular markers system. Seeds that were collected from different localities were planted in a controlled glasshouse within pots, DNAs were isolated from young leaf samples of the seedlings using 2 x CTAB procedure and genotyping was conducted by selected ISSR primers. Data were analyzed with NTSYS-pc 2.1 package program and polymorphisms of the populations were determined as 88.23%. In the study, genetic diversity of the chickpea gene resources belonging to Konya region was determined.

ISSR Markörleri Kullanarak Konya Bölgesinden Toplanan Nohut (Cicer arietinum L.) Popülasyonları Arasında Genetik Çeşitliliğin Belirlenmesi

Abstract

Bu çalışma, nohut yetiştiriciliğinde büyük bir paya sahip olan Konya’daki farklı lokalitelerden toplanan nohut popülasyonlarının sahip olduğu genetik çeşitliliğin tespit edilmesi için yapılmıştır. Ülkemiz açısından ekonomik değeri olan, bölgeye uygun nohut çeşitleri geliştirmeye yönelik ıslah programlarına katkıda bulunmak hedeflenmiştir. Çalışmada, Konya Bölgesi’nden toplanmış olan 23 nohut popülasyonu ve 2 nohut çeşidi (Gökçe, Er 99) kullanılmış ve basit dizi tekrarlar arası (ISSR) moleküler markör tekniği ile popülasyonların aralarındaki genetik uzaklıklar belirlenmiştir. Farklı yerlerden temin edilen tohumların ekimi saksılarda kontrollü sera şartlarında gerçekleştirilmiş, yetiştirilen fidelerden alınan genç yaprak örneklerinden 2x CTAB metoduna göre DNA’ları izole edilmiş ve seçilmiş ISSR primerleri ile yapılan PCR işlemleri sonucunda oluşan bantlardan popülasyonların ortalama polimorfizm oranı %88.23 olarak belirlenmiştir. Elde edilen veriler NTSYS-pc 2.1 programı ile analiz edilmiş ve genetik ilişki dendogramı oluşturulmuştur. Çalışma sonucunda Konya Bölgesine ait nohut gen kaynaklarının sahip olduğu genetik çeşitlilik moleküler markörler ile ortaya konmuştur.
