The Investigation of In vitro Regeneration System of Astragalus schizopterus

Abstract

In this study, in vitro germination and regeneration ability of endemic Astragalus schizopterus Boiss. was investigated. As we encounter germination handicap, the seeds of A. schizopterus plant,were subjected to sandpaper, boot with a scalpel, or hydrochloric acid (HCl) treatments. The highest germination rate (96.7%) was obtained from seeds plotted with a scalpel. Leaf and petiole explants taken from seedlings grown in aseptic conditions, were cultured in media supplemented with 2.0, 4.0, 8.0 mg/l 2,4-D x 0.5 mg/l KIN containing MS or B5 media, 0.0, 0.5, 1.0, 2.0, 5.0 mg/l 2,4-D x KIN containing MS media, 2.0, 4.0 mg/l IAAx0.5 mg/l and 0.5 mg/l ZEA, l.0 IAAx2.0, 4.0 mg/l ZEA containing MS nutrient environments, hypocotyl explants were cultured in 1.0, 2.0, 4.0 mg/l 2,4-Dx0.5 mg/l BAP or KIN in MS containing environments, ad-ventitious shoot regeneration or somatic embryos were cultured for stimulation. As a result, mostly soft or hard bodied and non-embryogenic callus tissues were obtained from the explants taken from A. schizopterus. There was no shoot regeneration from the callus in any environment, although root formation was observed in some environments.

Astragalus schizopterus’da In vitro Rejenerasyon Sisteminin Araştırılması

Abstract

Bu araştırmada, endemik Astragalus schizopterus Boiss.’in çimlenme ve in vitro rejenerasyon yeteneği araştırılmıştır. Karşılaşılan çimlenme probleminden dolayı A. schizopterus bitkisinin tohumları, zımparalama, bisturi ile çizme ve hidroklorik (HCl) asit ile muamele gibi uygulamalara tabi tutulmuştur. En iyi çimlenme oranı (% 96.7) bisturi ile çizilen tohumlardan elde edilmiştir. Aseptik şartlarda yetiştirilen fidelerden alınan yaprak ve yaprak sapı eksplantları 2.0, 4.0, 8.0 mg/l 2,4-Dx0.5 mg/l KIN içeren MS veya B5 ortamları, 0.0, 0.5, 1.0, 2.0, 5.0 mg/l 2,4-DxKIN içeren MS ortamları, 2.0, 4.0 mg/l IAAx0.5 mg/l ZEA ile 0.5 mg/l IAAx2.0, 4.0 mg/l ZEA içeren MS besin ortamlarında, hipokotil eksplantı 1.0, 2.0, 4.0 mg/l 2,4-Dx0.5 mg/l BAP veya KIN içeren MS ortamlarında, adventif sürgün rejenerasyonu veya somatik embriyo uyarımı için kültüre alınmıştır. Sonuç olarak A. schizopterus’ta denemeye alınan eksplantlardan çoğunlukla yumuşak veya sert yapılı ve embriyogenik olmayan kalluslar elde edilmiştir. Rejenerasyon ortamına alınan kalluslarda herhangi bir sürgün rejenerasyonu görülmezken, bazı ortamlarda kök oluşumu gözlenmiştir.
