Ergin ve Ergin Olmayan Gökkuşağı Alabalığı Oncorhynchus mykiss’in (Osteichthyes: Salmonidae) Karaciğer Lipaz Enzimi (E.C 3.1.1.3) Aktivitesinin Belirlenmesi
Abstract
Bu çalışmada, ergin ve ergin olmayan Oncorhynchus mykiss’ in karaciğer lipaz (E.C 3.1.1.3) enzimi aktivitesinin belirlenmesi amaçlanmıştır. Ergin ve ergin olmayan O. mykiss karaciğer lipazını saflaştırma işlemleri 3 adımdan oluşmuştur: Homojenatın hazırlanması, amonyum sülfat çöktürmesi, Diethylamino ethyl (DEAE) selülozik iyon değişim kromatografisi. Saflaştırma işlemleri boyunca, ergin balıkların karaciğerinden elde edilen lipaz enziminin spesifik aktivitesi 2.23±1.59 µmol/mg protein, verimi %0.037 ve saflaştırma katsayısı 2.65±1.94 olarak belirlenmiştir. Ergin olmayan balıklarda ise spesifik aktivite 1.86±0.14 µmol/mg protein, verim %3.24±0.51 ve saflaştırma katsayısı 15.50±1.13 bulunmuştur.
