Ulex europaeus Lektininin Kan Grubu Bağlama Özgüllüğü

Abstract

Bu çalışmada, Ulex europaeus bitkisinden elde edilen Ulex europaeus Aglutinin-I (UEA-I)’in insan kan grubu bağlama özgüllüğü ölçüldü. Bu deney için U tabanlı mikrotitrasyon kabı kullanıldı. 50µg/ml lektin çözeltisi, PBS (phosphate buffered saline) ile hazırlanarak başlangıç konsantrasyonu olarak alındı ve UEA-I’in seri sulandırılması ile insan kan grubu özgüllüğü belirlendi. Sonuç olarak, UEA-I’in %2’lik A grubu eritrositlerini 25µg/ml konsantrasyonda, B grubu eritrositlerini 6.25µg/ml konsantrasyonda, AB grubu eritrositlerini 50µg/ml konsantrasyonda ve O grubu eritrositlerini 1.57µg/ml konsantrasyonda aglutine ettiği belirlendi. Bu konsantrasyonlara bakılarak, UEA-I’in O grubu insan kanı eritrositlerine karşı spesifik olduğu tespit edildi.

Blood Group Binding Specificity of The Lectin, Ulex europaeus

Abstract

: In this study, it was measured the human blood group binding specificity of the Ulex europaeus Agglutinin-I (UEA-I) from Ulex europaeus. U-based microtitration plate was used for this experiment. 50µg/ml lectin solution was prepared with PBS and taken as a initiation concentration, and human blood group specificity of UEA-I was determined by use the serial dilution of lectin solution. As a result, it was observed that the UEA-I agglutinated the 2% A group erythrocytes at the concentration of 25µg/ml, the B group erythrocytes at the concentration of 6.25µg/ml, the AB group erythrocytes at the concentration of 50µg/ml and O group erythrocytes at the concentration of 1.57µg/ml. According to the results, it was determined that UEA-I is specific for O human blood group erythrocyte at that concentration examined.
