Bu çalışmada Pleurotus ostreatus’un misel gelişimi
üzerine sitokinin olarak 6N-benzil adenin (BA) ve kinetin (Kin)’in etkisi
araştırılmıştır. Besi yeri olarak malt ekstrakt agar (MEA) ve agar agar (AA)
kullanılmıştır. Malt ekstrakt agar ve agar agara sitokininlerin %0.1, 0.3 ve
0.5 konsantrasyonları ilave edilerek otoklavda steril hale getirilmiştir.
Ortamlar biyogüvenlik kabini içerisinde steril edilmiş petrilere dökülerek
katılaşmaları sağlanmıştır. Besi yerlerine misel ihtiva eden 1 cm çapında
kültür ilavesi yapılmış ve etüve alınmıştır. Belli zaman aralıklarıyla miselyum
gelişmeleri alansal olarak çizilmiştir. Çalışma, miselyumun petrinin toplam
alanını dolduruncaya kadar devam ettirilmiştir. Çalışma sonucunda her iki
besiyerindeki kontrol gruplarına göre, MEA+%0.5 Kin ile AA+%0.5 BA içeren
petrilerde diğer kombinasyonlara göre daha erken bir gelişme tespit edilmiştir.
MEA besiyerinde gelişme sık-yoğun iken AA besiyerinde zayıf-seyrek bir gelişme
gözlenmiştir.

In this study, the effect of 6-benzyl adenine (BA) and kinetin (Kin),
known as cytokinin, on the development of micelle of Pleurotus ostreatus was investigated. Malt extract agar (MEA) and
agar agar (AA) were used as a feedstock. By adding 0.1%, 0.3% ve 0.5% doses of cytokinins to MEA
and AA were sterilized in the autoclave. The nutrients were poured into the
pre-sterilized petri dishes in the biosafety cabinet and solidified. 1 cm
diameter mushroom culture was added to the feedstock and petri dishes were taken
to the incubator. The development of mycelium at certain time intervals has
been drawn. Work continued until the mycelium had filled the total surface area
of the feedstock. According to the experimental results, it was found that the
earlier development has been detected from the petri dishes containing with MEA
added with 0.5% Kin and AA added with 0.5% BA, compared to the control groups
on both feedstock. While growth in MEA medium was frequent-dense, weak-sparse
development was observed in AA medium.
