Pancreas Tissue Fixation Using Various Methods

Abstract

Fixation is the main step in the examination of the tissues by light microscopy and effects the following procedures. There are various fixation methods as well as various fixatives that can be utilized by these methods. The aim of this study is to define which fixative gives the best result in the examination of endocrin or exocrine parts of the pancreas. For this aim, two different fixation methods (perfusion and immersion), three different fixatives (neutral formaline, Bouin's fluid, 4% paraformaldehyde+7.5% picric acid) and different fixation times are applied onto rat pancreas tissues. Microwave irradiation is applied to some of the tissues which were fixed by either method. The sections examined were stained with heamatoxylin and eosin. We conclude that the results of the present study will guide the future studies which would be performed to analyze the endocrine or exocrine part of the pancreas.

Pankreas Dokusunun Değişik Yöntemlerle Fiksasyonu

Abstract

Tüm dokuların ışık mikroskopik incelemesinde fiksasyon temel basamaktır ve sonraki aşamaları etkiler. Değişik fiksasyon yöntemleri olduğu gibi, bu amaç için kullanılan farklı fiksatifler de bulunmaktadır. Bu çalışmada pankreasın gerek endokrin gerekse ekzokrin bölümünün inceleneceği çalışmalarda hangi fiksatifin seçilmesi ile iyi sonuç elde edilebileceğinin araştırılması planlanmıştır. Bu amaçla iki farklı fiksasyon yöntemi (perfüzyon ve immersiyon), üç farklı fiksatif (nötral formalin, Bouin sıvısı, %4 paraformaldehit+%7.5 pikrik asit) ve değişik fiksasyon süreleri sıçan pankreas dokularında denenmiştir. Her iki yöntemle de fikse edilen dokuların bir kısmına mikrodalga ışınımı uygulanmıştır. Elde edilen kesitler hematoksilen ve eozin ile boyanarak değerlendirilmiştir. Bu çalışma sonucunda elde edilen verilerin pankreasın ekzokrin ya da endokrin bölümünün incelenmesi amacı ile yapılacak çalışmalara yön vereceği düşüncesine varılmıştır.
