Flow Cytometric Analysis Of In Vitro Activation Markers Of Platelets Collected With Two Different Cell Separators

Abstract

The aim of this study was to compare in vitro activation markers of platelets collected with two different cell separators (Cobe Spectra and CS3000 plus) analysed by flow cytometry on day 0 and 3, to determine the relationship between platelet activation and storage time. 5O donor (5 women 45 men) aged between 20-49 (median age 33.4±8.7) who were good candidate for apheresis and who admitted to apheresis for the first time were selected for the study. The platelet activation markers of collections were assessed on day 0 and on day 3 by flow cytometry. The 0. day analysis was performed without time delay as soon as the concentrate was collected and 3. day analysis was performed on the product which was stored on ajitator. CD62 and CD63 expression percentage were high on the third day of storage in both collections of two cell separators Cobe Spectra and CS3000 plus.
