Roles of Sympathoadrenal System and Nitric Oxide In The Effect of Adrenomedullin that is Injected Intracerebroventricularly on Blood Pressure and Heart Rate

Abstract

In the study presented, we investigated the roles of sympathetic system and the central nitric oxide (NO) in the blood pressure and heart rate-increasing effects of adrenomedullin (ADM) applied intracerebroventricularly. The right lateral ventricles were cannulated under the ether anesthesia for measuring blood pressure of the right femoral artery of rats and for intracerebroventricular injections. To determine the role of sempathetic system in the increasing effects of ADM on blood pressure and heart rate, α-adrenoceptor antagonist phentolamine or β-adrenoceptor antagonist propranolol were administered before ADM. Moreover, intraperitoneal hexametonium was injected before ADM to identify the role of autonomic ganglionic blockage. Nitric oxide synthase inhibitor L-NAME was applied before ADM to find out the role of central NO in the effects of ADM. Phentholamine and propranolol prevented the cardiovascular effects of ADM. It is notable that the hypo-tension caused by hexametonium was partially recovered after ADM injection and L-NAME prevented the effects of ADM only on the blood pressure. Our findings revealed that the centrally applied ADM increased blood pressure and heart rate through leading to a sympathetic system stimulation via α and β adrenergic receptors. It is thought that the central NO could have a role in the central cardiovascular effects of ADM.
