Histologic Evaluation of Uterus During Rat Implantation Process

Abstract

Embryo implantation process following fertilization contains complex mechanisms as the most important stage for healthy pregnancy in humans. “Implantation window” is defined in humans as the 6-10th days after fertilization, whereas in rats it is defined as the 4.5-6.5th days by considering the similarities with human cycle. Implantation process have an important role in studies investigating the mechanisms that cause infertility. In this study, we aimed to create a new resource for new studies using histological evaluation in regard to the compilation of the recent literature. Female rats (n=21) divided into 3 groups as embryonic days of 4.5th, 5.5th and 6.5th according to the vaginal smear examinations which determined the estrous after fertilization. Uterus samples of defined groups of pregnant female rats were obtained. The samples fixed in 10% formaline solution and prepared according the routine paraffin tissue protocol and embedded in paraffin. Five-mikrometer-thick paraffin-embedded sections were stained with Hematoxylin-Eosin and Periodic acid Schiff. The basic structures were evaluated in the process of implantation in the stained uterus sections. Basic structures in implantation process as endometrium, myometrium, perimetrium, surface epithelium, primary and secondary decidual areas, GMG cells, trophoblast and embryoblast cells that have seen in sections were compatible within the groups. The results were compared between the stages of the implantation process. In this study, as one of the most important stage for healthy pregnancy, the embryo implantation process which contains a comlex mechanism was histologically evaluated in rats that were used as a model organism.

Sıçan İmplantasyon Dönemi Boyunca Uterusların Histolojik Değerlendirmesi

Abstract

İnsanlarda embriyo implantasyon süreci karmaşık fizyolojik mekanizmalar içermekte olup, sağlıklı gebeliğin elde edilmesi için fertilizasyo-nun ardından gerçekleşen en önemli aşamadır. İnsanlarda fertilizasyonun ardından 6-10. günler, insanlara benzer döngüleri nedeni ile model organizma olarak kullanılan sıçanlarda ise 4.5-6.5. günler “implantasyon penceresi” adı verilen süreci kapsamaktadır. İnfertiliteye neden olan mekanizmaların araştırıldığı çalışmalarda implantasyon süreci önemli rol oynamaktadır. Bu çalışmada sıçan implantasyon dönemi ile ilgili histolojik değerlendirmeler ve elde edilen son literatür bilgilerinin derlenmesi ile yeni çalışmalar için kaynak oluşturulması amaçlanmıştır. Vaginal yayma ile östrus evresinde belirlenen dişi sıçanlar, fertilizasyonun ardından; embriyonik gelişimin 4.5, 5.5. ve 6.5. günü olmak üzere 3 gruba ayrıldı ve grupları belirlenen dişi sıçanlardan (n:21) uterus örnekleri elde edildi. Dokular %10'luk formalinde tespit edilerek, rutin parafin doku takibi ile parafine gömüldü. Beş μm.'lik parafin kesitler alınarak Hematoksilen-Eozin ile boyandı ve Periyodik asit Shiff reak-siyonu ile değerlendirildi. Boyanmış kesitlerde implantasyon sürecinde uterustaki temel yapılar değerlendirildi. Kesitlerde implantasyon sürecinde görülen temel yapılar olan endometriyum, miyometriyum, perimetriyum, yüzey epiteli, primer ve sekonder desidual alanlar, GMB hücreleri, trofoblast ve embriyoblast hücreleri gösterilmiştir. Sonuçlar implantasyon dönemi evreleri boyunca karşılaştırılmıştır. Sağlıklı gebeliğin elde edilmesi için en önemli evrelerden biri olan implantasyon süreci model organizma olarak yaygın olarak kullanılan sıçanlarda evrelere göre değerlendirilmiştir.
