Effect of Different Combinations of BAP and NAA in vitro Propagation of Wild Safflower (Carthamus persicus Wild)
Abstract
Safflower (Carthamus persicus Wild), is rich in terms of linoleic acid and oleic acid and is used to obtain low saturated fatty acid. In order to propagate safflower in vitro, shoot tip explants were cultured in MS medium containing different combinations of BAP and NAA supplemented with 3% sucrose and solidified with 0.8% agar. The results showed a significant interaction between BAP and NAA combinations for callus formation, shoot and root regeneration, number of shoots and roots per explant, shoot and root length. Maximum shoot regeneration was noted in MS medium containing 0.50 mg/L BAP + 0.25 mg/L NAA. Maximum callus formation was determined in MS medium supplemented with 1 mg/L BAP + 1 mg/L NAA and 2 mg/L BAP + 1mg/L NAA. The regenerated shoots were rooted on MS medium containing 1 mg/L IBA.
