Propagation of GF 677 (P. amygdalus x P. persica) Clone Rootstock in Tissue Culture by Shoot-tip Techique

Abstract

 

Modified Murashige and Skoog (MS) nutrient medium was used to propagate the GF 677 clone rootstock by shoot tip technique.  Survival ratio of shoot tips was differed between 38% and 60% and increased by BAP concentration. The best multiplication was obtained on a MS medium supplemented with 2.0 mg/l BAP, 0.1 mg/l NAA and 0.1 mg/l GA3. In general multiplication ration was increased when GA3 added to medium. Rooting of in vitro plantlets was better with NAA than IBA while there was no significant difference between auxins. The highest rooting was obtained on half or full strength MS medium containing 1.0 and 1.5 mg/l NAA or IBA. Half or full strength MS medium were showed similar  affect on rooting.
