The evaluation of osteolysis resulting from titanium particles in -Interleukin-6 gene knocked out- transgenic mice

Abstract

Titanium and titanium alloys which are used for components of total joint replacement prosthesis have been shown to generate more wear debris. The cellular response to titanium is characterized by release of tumor necrosis factor (TNF) from macrophages and prostaglandin E2 (PGE2 ) from osteoblasts. The presence of numerous macrophages at the interface of a loose prosthesis may lead to increased levels of cytokines (i.e. TNF, interleukin-1, 6, 11). This study was designed to define the role titanium debris plays in osteolysis and to indicate the importance of interleukin-6 (IL-6) in this process. Twenty female transgenic mice were used for this purpose. They were divided into two groups. First group (10 mice) had IL-6 gene whereas second group hadn't ( IL-6 gene was knocked-out ). Five mice were used as control from each group. Commercially pure titanium particles were placed on calvarias of mice except controls. They all were sacrificed after one week and their calvarias were harvested. Investigation was done histomorphometrically. The group which had not IL-6 gene indicated significantly less osteolysis than the other group (p<0.05). We conclude that IL-6 had an important role in osteolysis resulting from titanium particles.
