Investigation of the Presence of Escherichia coli and Escherichia coli O157:H7 Serotype with Clinical and Hematological Findings in Calves with Diarrhea in Van and its Region

Abstract

The aim of this study is to investigate the clinical and hematological findings and the presence of Escherichia coli (E.coli) and E.coli O157:H7 serotype in calves with diarrhea in Van and its region. In the present study a total of 74 calves aged between 1 and 30 days in different breeds were tested. Out of 74 calves, 54 were diarrheic and 20 were healthy. Escherichia coli (E. coli) O157:H7 serotype together with some hametological parameters (Red blood cell (RBC), White blood cell (WBC), Hemoglobin (Hb), Packed cell volüme (PCV)) were investigated in the faeces of these diarrheic and healthy calves. According to examination and evaluation protocol, out of 54 diarrheic calves, 20 had a mild dehydration, 20 had medium dehydration and 14 had severe dehydration. Haematolojic parameters in calves with mild dehydration were not significantly different compared to the values obtained from control animals. However, PCV and Hb values were found to be significantly different (p<0.001) in calves with medium and severe dehydration. RBC in calves with medium (p<0.01) and severe (p<0.001) dehidration were also significant compared to control values obtained from healthy calves. Furthermore, the number of WBC were significantly (p<0.05) different both in calves with medium and severe dehydration. Although E.coli determined 74,6% in the diarrhoeic calves after microbiological analysis, E.coli O157:H7 serotype couldn’t be isolated. As a result, important changes in RBC, WBC, PCV and Hb values obseved in calves with diarrhea and E.coli was one of the most important causative agent calf diarrhea. It was also determined that E.coli O157:H7 serotype isn’t a pathogen seen very often.

Van ve Yöresinde İshalli Buzağılarda Klinik ve Hematolojik Bulgular ile Escherichia coli ve Escherichia coli O157:H7 Serotipin Varlığının Araştırılması

Öz

Bu çalışmanın amacı Van ve Yöresindeki ishalli buzağılarda klinik ve hematolojik bulgular ile Escherichia coli (E.coli) ve E.coli O157:H7 serotipinin varlığını araştırmaktır. Araştırmanın materyalini, 1-30 günlük yaşlarda değişik ırk ve cinsiyette 54 ishalli ve 20 sağlıklı (kontrol grubu) olmak üzere 74 buzağı oluşturdu. Buzağılarda, kan eritrosit (RBC), lökosit (WBC), hemoglobin (Hb), hematokrit (PCV) değerlerine, dışkıda E.coli ve E.coli O157:H7 serotipine bakıldı. Klinik muayaneleri yapılan 54 buzağının 20 sinin hafif, 20’sinin orta ve 14’ünün ise şiddetli derecede dehidre olduğu belirlendi. RBC, WBC, PCV ve Hb değerleri hafif derecede dehidre buzağılarda, sağlıklı olan kontrol grubu ile kıyaslandığında istatistiki olarak önemsiz bulunmuştur. Orta ve şiddetli derecede dehidre buzağılarda PCV ve Hb değerleri, kontrol grubu ile kıyaslandığında, istatistiki olarak çok önemli fark belirlenmiştir (p<0.001). Orta derecede dehidre buzağılarda RBC düzeyi kontrol grubu ile kıyaslandığında önemli derecede farklılık tespit edilirken (p<0.01), şiddetli derecede dehidre buzağılarda ise istatistiki olarak çok önemli düzeyde (p<0.001) farklılık tespit edilmiştir. WBC düzeyi ise hem orta hem de şiddetli derecede dehidre buzağılarda, kontrol grubu ile kıyaslandığında önemli düzeyde (p<0.05) farklılık bulunmuştur. İshalli buzağıların dışkı örneklerinin mikrobiyolojik incelenmesinde %74.6 oranında E.coli izole edilmesine rağmen E.coli O157:H7 serotipi izole edilemedi. Sonuç olarak buzağı ishallerinde kanda RBC, WBC, PCV ve Hb değerlerinde önemli değişikliklerin olduğu, buzağı ishallerinde E.coli’nin önemli bir etken olduğu ancak E.coli O157:H7 seropitinin sık rastlanan bir patojen olmadığı belirlendi.
